The present disclosure provides fluorogenic compounds useful for preparing fluorescent calcium ion indicators, the fluorescent indicators themselves, and the use of the fluorescent indicators in methods of detection, discrimination and quantification of metal ions. The subject fluorogenic compounds and fluorescent ion indicators can include a chelating group based on a 2-aminophenoxyethylene glycol 2-aminoethyl ether, N,N,N',N&#x2032;-tetraacetic acid (PEGTA) moiety or precursor thereof where the phenyl group of the PEGTA is substituted with or fused with a fluorophore moiety of interest. The subject methods find use in the detection of intracellular calcium ions. Also provided are kits for use in practicing the subject methods.

Chemically reactive dyes that are intramolecularly crosslinked with a water-soluble bridge, their bioconjugates and their uses are described. Reactive fluorescent dyes that have a water-soluble bridge are superior to those of conjugates of spectrally non-crosslinked dyes or the dyes that are crosslinked with a hydrophobic bridge. The invention includes reactive fluorescent dyes, their biological conjugates and uses.

Luminescent dyes with a water-soluble intramolecular bridge and their biological conjugates

Chemically-reactive, water-soluble, heterocycle-substituted 7-hydroxycoumarin dyes, their bioconjugates and uses are described. The conjugates derived from reactive heterocycle-substituted 7-hydroxycoumarin dyes are used for analyzing biological compounds. These heterocycle-substituted 7-hydroxycoumarin dyes are particularly useful as fluorescent labels for biopolymer detection reagents, such as antibodies or nucleic acid probes. The dye-antibody conjugates of the invention are particularly useful for analyzing analytes using a flow cytometer equipped with a violet laser as an excitation source due to their strong absorption at 405 nm and high fluorescence quantum yield.
