Intraoperative optoacoustic guide apparatus and method

A method for the determination of antibiotic susceptibility through stimulated Raman scattering microscopy is disclosed. The method utilizes a imaging apparatus adapted to collect a laser signal through a sample having a bacteria for imaging the metabolism of the sample. The sample can be manipulated with an antibiotic for imaging to determine the susceptibility of a bacteria to the provided antibiotic.

A photoacoustic catheter includes an elongated catheter body and a housing positioned near a distal end of the elongated catheter body. A length of multimode fiber extends through the elongated catheter body and has a distal end that is beveled at about 45&#xb0; relative to a longitudinal axis of the multimode fiber and is positioned in the housing. An ultrasonic transducer, electrically connected to an electrical wire extending along the elongated catheter body, is positioned within the housing. A mirror element is also positioned within the housing and includes a mirror surface beveled at about 45&#xb0; relative to the longitudinal axis of the multimode fiber. The catheter is operable to deliver an optical wave through the multimode fiber and to deliver an ultrasonic wave collinearly from the housing and out of an aperture of the housing to obtain optical data and ultrasonic data within a mammalian luminal organ.

A method of noninvasively imaging tissue within a body includes irradiating the tissue using an imaging laser including a Raman-based laser tuner, the radiation including a plurality of laser pulses, each having energy greater than 100 mJ; receiving an acoustic signal generated by vibrational energy in the tissue, wherein the vibrational energy is a result of selective overtone excitation of molecules in the tissue by the radiation; and automatically converting the acoustic signal to an image representative of the tissue using a processor. An imaging system includes an imaging laser configured to irradiate tissue with a plurality of laser pulses using a Raman-based laser tuner. An ultrasonic transducer receives an acoustic signal generated by vibrational energy in the tissue due to overtone excitation by the radiation. A processor is configured to automatically produce an image representative of the tissue using the received acoustic signal.

Vibrational photoacoustic tomography using Raman laser

An optical imaging apparatus includes an optical signal source, an optical signal detector apparatus, and a resonant amplifier assembly. The optical signal source is configured (i) to generate an optical signal including a carrier signal and an imaging signal, and (ii) to guide the optical signal to a sample. The optical signal detector apparatus is configured (i) to detect a modified optical signal from the sample, and (ii) to generate an electrical image signal based on the modified optical signal. The electrical image signal includes a background component and a modulated image signal corresponding to an image of the sample. The invention generally relates to methods for determining aggressiveness of a cancer and treatment thereof. Certain aspects of the invention provide methods that involve conducting an assay on a lipid droplet in order to detect an amount of a biomarker within the lipid droplet, and determining aggressiveness of a cancer based upon the amount of the biomarker within the lipid droplet. Other aspects of the invention provide methods for treating a cancer that involve administering an agent that blocks storage of cholesteryl ester in a lipid droplet.

A system for measuring an sample includes an illumination source providing electromagnetic radiation pulses at a selected temporal frequency. A microscope focuses the radiation to interact with the sample and produce resultant electromagnetic radiation. Optical detection apparatus includes the optical sensors, resonant amplifiers, and disperser. A method of analyzing constituents of a sample includes contemporaneously irradiating the sample with narrowband light and broadband light, the optical power of either modulated at a selected temporal frequency; dispersing wavelengths of resultant light across the optical detectors; and filtering respective signals from the optical detectors to provide spectrum data including signals corresponding to the selected temporal frequency.

System and method for multiplex spectroscopic imaging

A method for providing images using a multimodal nonlinear optical microscope is disclosed. The method includes providing a foundation femtosecond laser beam, generating a first femtosecond laser beam and a second femtosecond laser beam corresponding to the foundation femtosecond laser beam, combining the first femtosecond laser beam and the foundation femtosecond laser beam to generate a first combination femtosecond laser beam, and generating a coherent anti-Stokes Raman scattering (CARS) signal based on the first combination femtosecond laser beam.
