An approach is described that combines distinct properties of a specialized porous nitrocellulose film (PNC) with quantum nanoparticles to create an improved assay and detection sensitivity, permitting the development of a camera-based imaging system for fluorescent detection of macromolecules in microarray format. The two properties of PNC that facilitate the approach are an extraordinarily high binding capacity and a newly observed internal scattering of light. Quantum nanoparticles complement these PNC properties by providing a higher level of emitted light than the fluorescent dyes in common microarray use. Overall, the approach allows for instrument cost savings, reduced imaging time, and the ability to remotely image.

Protein microarray assay imager

Methods and systems are provided for imaging a microarray of a microarray assembly. The microarray assembly may be configured as a protein microarray and may be used to visualize protein expression levels in sample for disease detection and/or diagnosis. In one example, the microarray assembly may comprise a laser pointed in a first direction, a camera positioned parallel to and vertically below the laser, a first dichroic mirror vertically aligned with the laser for reflecting light emitted from the laser, a second mirror vertically aligned with the camera and horizontally aligned with the first dichroic mirror, and a chip coated in a nitrocellulose film and including an array of wells containing one or more biomolecules.

Portable microarray assembly

Methods and systems are provided for a lateral flow test device. In one example a lateral flow test device may include a housing comprising an upper first portion and a lower second portion, the lower second portion further including a planar surface, a nitrocellulose matrix strip, the strip disposed on the planar surface, and one or more ligand regions included in the strip, the ligand regions comprising one or more ligands. The strip may be formed from a liquid polymer mixture dispensed onto the planar surface via a dispensing device positioned vertically above the planar surface.

A reaction surface array diagnostic apparatus and method of making the same includes a substrate carrying a plurality of reaction surfaces, a plate and a gasket, each having a plurality of through bores, alignable with one of the reaction surfaces and forming a fluid tight well about each reaction surface when the gasket and the plate are sealingly affixed to the substrate to form a stack. Clamp members engage opposite side edges of a stack to compress the gasket. A plurality of side-by-side disposed clamped stacks of plates, gaskets and substrates are mounted in a tray in the standard footprint of a microtitre plate. Alternately, the plate and the gasket are combined into a single plate formed of a flexible material having an adhesive on one surface.

Reaction surface array diagnostic apparatus

A reaction surface array diagnostic apparatus includes a substrate carrying a plurality of reaction surfaces and, a plate having a plurality of wells, alignable with one of the reaction surfaces and forming a fluid tight well about each reaction surface when the plate and the substrate are sealingly affixed to each other in a frame. The plate is formed of a flexible material which may have an adhesive on one surface. Interlocking latch members are formed on the plate and the frame for releasably latching the plate and the frame in a position for sealing attachment to the substrate.

The invention describes a process for preparing a microtiter-microarray device that includes a bottomless microtiter plate attached to the first side of one or more substrates with microarrays of materials attached thereto. The microtiter plate and the one or more substrates are attached through one or more gaskets. Preferably, the microtiter plate is attached to one face of the one or more gaskets by an irreversible water-tight seal, and the first side of the one or more substrates is attached to the opposite face of the one or more gaskets by a reversible, water-tight seal.

Interface between substrates having microarrays and microtiter plates

A probe clip for in situ assay of tissue sections in the form of a plate having a first seal member mounted thereon and forming an interior cavity on the plate. In one embodiment, a second seal member is mounted interiorly of the first seal member and divides the interior cavity into first and second fluid communicatable surfaces, with a probe dryingly attached to the plate and disposed on the second mixing surface. The plate is joined to a slide carrying a tissue section and a reactant fluid such to form fluid communicatable reaction and mixing chambers. Successive rotations of the joined plate and slide causes the reactant fluid to initially flow to the mixing chamber and release the probe, before the probe flows to the reaction chamber for reaction with the tissue section. In another embodiment, a time-release material covers the probe mounted on a plate having a single chamber. The reactant fluid hydrolyzes the time-release material to release the probe for reaction with the tissue. The cassette carrying one or more plates is slidingly insertable into a semi-closed housing containing one or more tissue-carrying slides. Clamps urge the probe clip cassette and the individual plates into engagement with the slides to form the sealed chambers therebetween. Inlet and outlet wash ports communicate with the slides to wash the slides after the plates have been removed from the housing.
