The gene encoding the ligninase rLDM.TM. 6 has been cloned and expressed. The ligninases produced via the processes disclosed herein are useful to degrade or modify lignin in pulp operations.

Cloning and expression of ligninases

The subject invention concerns a novel and superior recombinant DNA expression/secretion system for use in prokaryotic organisms to produce useful proteins. The system can be used to have proteins secreted by an engineered prokaryotic host where normally the proteins are not secreted by the host. Also, when using the expression system, proteins are expressed at much higher levels than when the expression system is not used.

Novel prokaryotic expression and secretion system

The invention concerns novel and useful mutant microbes which are capable of releasing substantial amounts of any of several periplasmic or recombinant proteins into the culture medium when carrying an expressed Kil gene. Though E. coli are exemplified, the invention is broadly applicable to the making of mutants of other microbes, for example, Salmonella, Klebsiella, and Rhizobium. A key feature of the invention is the use of a novel selection procedure employing a plasmid comprising the kil gene.
