A minimally invasive penetrating microelectrode array is used to generate localized electric field 'hotspots' for delivering biomolecules, such as nucleic acid or protein molecules, into cells located in the epidermal or dermal layer of the skin via transient membrane permeabilization. The “hotspots” can be controlled by selectively insulating the penetrating microelectrodes at specific regions. The portion of microelectrodes that are not covered with insulation coating can be coated with nucleic acid or protein vaccine vector, or other biomolecules to be delivered. Upon insertion into the skin, an anchor microelectrode region mechanically anchors the penetrating microelectrode to position the target tissue microelectrode region, so as to selectively align the biomolecule coating with cells located in the tissue location. The biomolecule coating will dissolve when in contact with surrounding tissue. By applying an electrical pulse, the biomolecules can be delivered into surrounding cells.

Device for tissue electrotransfer using a microelectrode

A method and apparatus for promoting the delivery of a molecule across a cell membrane comprises delivering a molecule to a delivery site of a region of tissue surrounding the cell membrane. A suction component applies a suction through a suction tip that surrounds the surface of the delivery site creating a seal to promote delivery of the molecule across the cell membrane. The application of the suction is controlled to create a predetermined negative pressure for a predetermined period of time before releasing the negative pressure. Suction is used to enhance permeabilization and transfection of cells. The technique can be used to enhance the uptake and subsequent transfection and expression of plasmid DNA vaccines, mRNA vaccines and other nucleic acid molecules, that are introduced subcutaneously. It can be used in combination with coated microneedles.

Provided is a fluidic device including a main channel, wherein a first inlet fluidly connects to an upstream end of the main channel and introduces magnetic beads into a first side of the main channel. A second inlet is fluidly connected to the upstream end of the main channel and introduces a sample stream into a second side of the main channel. A magnet disposed adjacent to the second side of the main channel pulls the magnetic beads towards a sidewall of the second side, and thus into the sample stream. The beads continue through an extended incubation channel before entering a return channel. The return channel includes a detection region. Also provided is a multi-layer micro-fluidic assay device. An assay method that utilizes a microfluidic assay device is provided as well.

A microelectrode array having a substrate with a plurality of grooves and a plurality of electrical contact pads, the grooves each with at least one electrode electrically connected to at least one of the electrical contact pads, each of the grooves containing at least one myotube that overlays the at least one electrode.
