A system and associated method for concentrating and separating components of different densities from fluid containing cells using a centrifuge includes a container defining a cavity for receiving the fluid. The container has a top, a sidewall extending from the top, and a bottom disposed opposite the top and in sealing engagement with the sidewall. An insert is slidably disposed in the cavity of the container and defines a lumen through the insert. The lumen, which includes a hole and a funnel-shaped upper portion in fluid communication with the hole, forms an open fluid path between opposite ends of the insert. The insert has a density such that upon centrifugation a selected component of the fluid resides within the lumen. A container port is disposed in the top of the container to transfer the fluid into the container and to withdraw a fluid component other than the selected component from the container. The system includes a manifold that includes a manifold port, a vent to vent the container, and a connector to couple to the container port. A cannula is receivable in the manifold port and extendable through the container port into the container and into the lumen of the insert to withdraw the selected component from the lumen.

A bone marrow aspiration device includes an introducer needle assembly and an aspiration needle assembly. The introducer needle assembly includes an introducer cannula and a removable trocar. The aspiration needle assembly includes a flexible aspiration cannula and a flexible stylet. The length of the aspiration cannula is substantially greater than the length of the introducer cannula. The aspiration needle assembly is receivable in the introducer cannula when the trocar is removed from the introducer needle assembly. The aspiration cannula forms a channel for aspirating bone marrow when the stylet is removed.

The present invention relates to antibodies with pH dependent binding to its antigen such that the affinity for antigen binding at physiological pH (i.e., pH 7.4) is greater than at endosomal pH (i.e., pH 6.0 or 5.5). In other words, the Kor kratio at pH 5.5/pH 7.4 or at pH 6.0/pH 7.4 is more than, or ranges between, 2, 3, 4, 8, 10, 16, 20, 30, 40, or 100 or more. Such pH dependent antibodies preferentially dissociate from the antigen in the endosome. This can increase antibody half life, as compared to antibodies with equivalent Ks at pH 7.4 but with no pH dependent binding, when the antigen is one that undergoes antigen-mediated clearance (e.g., PCSK9). Antibodies with pH dependent binding can decrease total antigen half life when the antigen undergoes reduced clearance when bound to antibody (e.g., IL6). Antibodies with pH dependent binding can also prolong the decrease in antigen which is not antibody-bound. This can be important when antagonizing a target antigen typically present at high levels (e.g., IgE, DKK1, C5 and SOST). In addition, such antibodies can increase antigen half life when the antigen is a receptor and the receptor has increased clearance when bound to antibody (e.g., GMCSF receptor).

Disclosed herein are apparatus and methods for isolating a fraction of interest from a physiological fluid sample. A sample holder for isolating a fraction of interest from a physiological fluid sample includes a flexible compartment and a rigid exoskeleton that supports the flexible compartment. The flexible compartment may have at least one reservoir with a height to volume ratio of about 0.1 cm/mL to about 5 cm/mL. An automated device for extracting a fraction of interest from the sample includes a sample holder with a flexible compartment supported by a rigid exoskeleton, a support for the sample holder connected to one or more fluid extraction devices, and a motor for moving the extraction device relative to the sample holder. The automated device may include an optical sensor and may include a clamp for clamping the flexible compartment.
