Described are methods of targeting specific genes or DNA sequences encoding biologically active proteins to the mammary gland such that the DNA sequence is expressed in the mammary gland and the protein is secreted into the milk of the mammal. Also described is the production of transgenic nonhuman mammals whose germ line comprises a DNA gene complex containing a casein promoter, a casein enhancer, a DNA sequence which encodes a signal peptide and a DNA sequence encoding a biologically active protein. The milk containing the biologically active protein or the biologically active protein itself can be used in the making of diary products.

Targeted synthesis of protein in mammary gland of a non-human transgenic

Several lines of transgenic mice have been generated using the prostate-specific rat probasin (PB) gene promoter to drive expression of the SV40 T antigen (Tag) coding region. Mice expressing high levels of the transgene specifically display progressive forms of prostatic cancer that histologically resemble human prostate cancer ranging from mild prostatic intraepithelial neoplasia (PIN) to large multinodular malignant adenocarcinoma with metastasis. Adenocarcinomas of the prostate have been detected as early as 10 weeks of age with extracapsular extension (ECE) and seminal vesicle invasion (SVI). Immunohistochemical analysis of tumor tissue has demonstrated that dorsolateral prostate-specific secretory proteins were confined to well differentiated ductal epithelial cells adjacent to, or within the poorly differentiated tumor mass. Prostate tumors in the mice also display elevated levels of nuclear p53, and a decreased heterogenous pattern of androgen receptor (AR) expression as observed in advanced human prostate cancer. The establishment of breeding lines of transgenic mice that reproducibly develop prostate cancer with metastasis provides a unique animal model system to study the molecular basis of transformation of normal prostatic cells and the factors influencing the progression to metastatic prostate cancer.

Transgenic mouse model for prostate cancer

Described is a method of targeting specific genes to the mammary gland which results in the efficient synthesis and secretion of biologically important molecules. Further, there is described as a composition of matter, a transgenic mammal having the ability to reproduce itself and being suitable for the secretion of biologically active agents into its milk. Additionally there is disclosed as a composition of matter, recombinant DNA gene complexes designed to integrate into a mammalian genome and to synthesize and secrete biological active agents into the milk. Furthermore methods of producing and using altered milk are disclosed.

DNA sequences to target proteins to the mammary gland for efficient

The device for retaining a slide and a pick to a stringed instrument includes a generally rectangular base having a top surface, and an arm unit extending over the base top surface. The arm unit includes an obliquely oriented slot arranged to accept and retain the pick therein. A gripping member extending from the arm unit forms a slide accepting area between the gripping member and the base top surface. A perimeter wall of the slide is inserted into the slide accepting area allowing the gripping member to apply a compressive force to the slide, retaining the slide to the device.
