A cell-based device and method are disclosed for in vitro testing of compositions or compounds for irritation potential. Both the device and method comprise a) a confluent layer of corneal epithelial cells supported on a chemically patterned hydrogel, where the cells are aligned with the chemical pattern, b) a collector to receive effluent fluid, and optionally c) a detector. The device and method can also comprise an extracellular matrix (ECM) assembled by corneal keratocytes supported on the chemically patterned hydrogel. The cell-based device and method provide simple replacements for the in vivo Draize rabbit eye irritation test.

In vitro cell-based replacement for the Draize test

Polymeric substrates, optionally in sheet form, treated with a thin layer of photoresist are perforated by laser ablation. Following removal of the photoresist the polymer substrates perforated with holes are patterned in stripes by photolithography, which is followed by synthesis of a cell-adhesive organometallic/self-assembled monolayer of phosphonate (SAMP) interface in the exposed regions, providing well-aligned continuous stripes for various levels of perforation. Cells plated on each of these 2-dimensional (2D) perforated surfaces attach to the interface and spread in alignment with pattern fidelity that is as high as that measured on a non-perforated, patterned substrate. A stack of such 2D patterned polymers yields a 3-dimensional (3D) device which facilitates cell growth and viability via the perforations.

Three-dimensional (3D) tissue scaffold with cell alignment

A method is disclosed for coating and patterning hydrogels in order to modify surface properties. The method exploits the water content of the hydrogel and the hydrophobicity of the reaction solvent to create a thin oxide adhesion layer on the hydrogel surface. This oxide adhesion layer enables rapid transformation of the hydrophilic, cell non-adhesive hydrogel into either a highly hydrophobic or a cell-adhesive hydrogel by reaction with an alkylphosphonic acid or an &#x3b1;,&#x3c9;-diphosphonoalkane, respectively. Also disclosed are coated, patterned hydrogels and constructs comprising the coated, patterned hydrogels.

A construct that supports cell attachment and alignment including a substrate that is incompatible with photolithography conditions, containing a physical pattern in at least part of one surface, the physical pattern optionally bearing a coating of a metal alkoxide, oxide or mixed oxide-alkoxide thereon and a Self-Assembled Monolayer of Phosphonate (SAMP) covalently attached thereto, which phosphonate contains functionality adapted for cell binding. The construct optionally also contains cells attached thereto. Also disclosed are methods of preparing such a construct.

Patterning of fragile or non-planar surfaces for cell alignment

Devices are provided which are functionalized to include surface regions having anti-infective agents. Methods are provided for functionalizing various material surfaces to include active surface regions for binding anti-infective agents. Methods are provided by which anti-infective moieties or agents are bonded to functionalized surfaces.

High-yield activation of polymer surfaces for covalent attachment of molecules

Phosphorus-based coatings having a plurality of phosphate moieties, a plurality of phosphonate moieties, or both, covalently bonded to an oxide surface of an implantable substrate exhibiting one or more of the following characteristics: (a) the surface phosphorus-containing group density of the coated regions of the substrate is at least about 0.1 nmol/cm; (b) the phosphorus-based coating has a thickness of less than about 10 nm; or (c) the surface phosphorus-containing group density of the coated regions of the substrate is equal to or greater than the surface hydroxyl group density of the oxide surface of the substrate. Implantable devices embodying the coated substrates are also disclosed.

Devices with multiple surface functionality

The present invention provides a dense-coverage, adherent phosphorous-based coating on the native oxide surface of a material. Disclosed phosphorous-based coatings include phosphate and organo-phosphonate coatings. The present invention also provides further derivatization of the phosphorous-based coatings to yield dense surface coverage of chemically reactive coatings and osetoblast adhesion-promoting and proliferation-promoting coatings on the native oxide surface of a titanium material.

Enhanced bonding layers on titanium materials

The present invention relates to a novel composition comprising an implant, scaffold or construct bound to a biological or chemical moiety. The bound moiety has the ability to bind to a component of the extracellular matrix of biological tissue, allowing the implant to be bound to the biological tissue in a short period of time after implantation. The invention also relates to the use and manufacture of this novel composition, as well as a novel use for the protein CNA.

Composition for the attachment of implants to collagen or other components of biological tissue

Nanoparticles are functionalized for use as bio-imaging probes using a novel, modular approach. Particle surface modification is based on a phosphonate monolayer platform on which was built a multi-segmented, multi-functional film: the first segment provided hydrolytic stability, the second aqueous suspendability, and the third, selectivity for cell attachment. In vitro imaging experiments visualized nanoparticle-cell surface binding. Peptide-derivatized nano-particles were not displaced from cells by soluble peptide. Methods for coating the host particles and use of rare earth ion-doped particles in imaging methods and photodynamic therapy methods are also disclosed.

Modular monolayer coatings for selective attachment of nanoparticles to biomolecules

A medical dressing for an open wound includes a base, and a raised flexible therapeutic cover formed with base. The base has opposing inside and outside major surfaces, and continuous inside and outside peripheral edges. The inside peripheral edge defines a wound site opening and designed to surround the open wound. The raised-flexible therapeutic cover extends over and above the wound site opening defined by the inside peripheral edge. A compression spring resiliently spaces the therapeutic cover from the open wound.
