Non-human animal genomes, non-human animal cells, and non-human animals comprising a humanized TRKB locus and methods of making and using such non-human animal genomes, non-human animal cells, and non-human animals are provided. Non-human animal cells or non-human animals comprising a humanized TRKB locus express a human TRKB protein or a chimeric transthyretin protein, fragments of which are from human TRKB. Methods are provided for using such non-human animals comprising a humanized TRKB locus to assess in vivo efficacy of human-TRKB-targeting reagents such as nuclease agents designed to target human TRKB.

Compositions and methods are provided for making rat pluripotent and totipotent cells, including rat embryonic stem (ES) cells. Compositions and methods for improving efficiency or frequency of germline transmission of genetic modifications in rats are provided. Such methods and compositions comprise an in vitro culture comprising a feeder cell layer and a population of rat ES cells or a rat ES cell line, wherein the in vitro culture conditions maintain pluripotency of the ES cell and comprises a media having mouse leukemia inhibitory factor (LIF) or an active variant or fragment thereof. Various methods of establishing such rat ES cell lines are further provided. Methods of selecting genetically modified rat ES cells are also provided, along with various methods to generate a transgenic rat from the genetically modified rat ES cells provided herein. Various kits and articles of manufacture are further provided.

Genetic modification of rats

Compositions and methods are provided for modifying a rat genomic locus of interest using a large targeting vector (LTVEC) comprising various endogenous or exogenous nucleic acid sequences as described herein. Compositions and methods for generating a genetically modified rat comprising one or more targeted genetic modifications in their germline are also provided. Compositions and methods are provided which comprise a genetically modified rat or rat cell comprising a targeted genetic modification in the rat interleukin-2 receptor gamma locus, the rat ApoE locus, the rat Rag2 locus, the rat Rag1 locus and/or the rat Rag2/Rag1 locus. The various methods and compositions provided herein allows for these modified loci to be transmitted through the germline.

A kitchen utensil or tool that includes a cutter, a hook, and a squeegee and that is particularly usable for interaction with bulk single use bags. The utensil is defined by a generally elongate body such that the hook and the cutter are oriented at a common end thereof. The body defines a handle that is associated with the opposite longitudinal end portion of the body. The body includes a slot that extends in the longitudinal direction such that the squeegee is defined by portions of the body adjacent the slot. An open end of the slot is disposed proximate the handle such that, during emptying of an opened bag, the hand of the user associated with the handle can support a cantilevered portion of the body that defines one of the sides of the squeegee.

A bolt hole marker for marking the location where a hole needs to be drilled in a foundation. The bolt hole marker includes an elongate angled base member having a first elongate portion having a slot in an end thereof with the slot being adapted to receive an anchor bolt, and further having a second elongate portion also having a slot in an end thereof with that slot being adapted to receive an anchor bolt. The second elongate portion is angled at approximately 30 degrees from the first elongate portion. A first punch member for making indentations in a sill member is disposed through the first elongate portion and is positioned approximately 3&frac14; inches from the slot of the first elongate portion. A second punch member for making indentations in a sill member is disposed through the second elongate portion and is positioned approximately 3&frac14; inches from the slot of the second elongate portion.
