Methods of detecting an analyte in a sample are provided. In one embodiment, the method comprises forming a partition comprising the sample, a binding agent capable of emitting a signal when bound to the analyte, and a marker capable of identifying the partition; allowing the binding agent to bind to the analyte, if present; and detecting the presence of the analyte in the sample by detecting the signal emitted from the binding agent in the partition.

Digital immunoassay

Methods of determining polymerase fidelity are provided. In one embodiment, the method comprises filling a gapped plasmid with a polymerase to form a gap-filled plasmid, wherein the gap-filled plasmid comprises a gene encoding an protein that is functional or non-functional depending on the polymerase fidelity; forming a plurality of partitions from a solution comprising the gap-filled plasmid and a label for detecting the presence of the plasmid; detecting the presence of the gap-filled plasmid in one or more of the partitions; and determining the fidelity of the polymerase by determining a ratio of partitions containing the gene encoding a functional protein to partitions containing a gene encoding a non-functional protein.

Digital polymerase fidelity assay

Sample analysis systems and methods are provided. In one embodiment, the method may be achieved by applying a substance to a surface of a substrate having a first binding agent immobilized thereon; removing unbound material from at least a portion of the substrate having the substance applied thereon; applying a second binding agent to the surface of the substrate, wherein the second binding agent is optically labeled or unlabeled; removing unbound material from at least a portion of the substrate having the second binding agent applied thereon; responsive to detecting the optically labeled second binding agent bound to the substance, identifying the analyte present in the sample; and responsive to not detecting the optically labeled second binding agent bound to the substance, determining that the analyte is absent in the sample; wherein the applying the substance or second binding agent to the surface of the substrate steps are concurrent with the respective removing unbound material from at least a portion of the substrate steps. Systems and other methods are also described and illustrated.
