The present invention concerns a detergent and a pharmaceutical composition comprising a deoxyribonuclease (DNase), wherein the DNase is obtained from a fungal source. It further concerns a laundering method and the use of DNase. The present invention further relates to polypeptides having DNase activity, nucleotides encoding the polypeptide, as well as methods of producing the polypeptides.

Detergent composition

The present invention relates to a recombinant fungal host cell comprising at least one first polynucleotide encoding a polypeptide of interest; and one or more second polynucleotide encoding a fungal PepC protease, wherein the one or more second polynucleotide is operably linked to a regulated heterologous promoter, as well as a method for producing a polypeptide of interest, comprising cultivating said fungal host cell.

Regulated PepC expression

The present invention relates to an isolated polypeptide having PepC inhibitory activity as well as to a method for producing a heterologous polypeptide of interest in anhost cell comprising: (a) cultivating thehost cell comprising a first and a second nucleic acid sequences under conditions conducive for the expression of the polypeptides encoded by the said first and second nucleic acid sequences, and wherein the first nucleic acid sequence encodes a heterologous polypeptide of interest and the second nucleic acid encodes the inhibitor polypeptide of the invention, and wherein the inhibitor polypeptide is expressed from a recombinant nucleic acid construct resulting in an increased level of the inhibitor polypeptide compared to anhost cell not comprising the recombinant nucleic acid construct; and (b) recovering the heterologous polypeptide.

Fungal PepC inhibitor

The disclosure relates to a filamentous fungal cell in which an endogenous alkaline protease activity, an endogenous neutral metalloprotease activity, an endogenous serine protease activity, and an endogenous kexin maturase activity have been completely or partially inactivated. Particularly the endogenous alkaline protease activity, the endogenous.neutral metalloprotease activity, the endogenous serine protease activity, and the kexin maturase activity are encoded by the alp, npI, pepC and kexB genes respectively. The filamentous fungal cell is particularly suitable for production of heterologous proteins such as antibodies.

Use of fungal mutants for expression of antibodies

The present invention relates to methods for producing a polypeptide of interest by (a) cultivating a mutant of a parentcell, wherein (i) the mutant comprises a first nucleic acid sequence encoding the polypeptide and a second nucleic acid sequence comprising a modification of at least one of the genes responsible for the biosynthesis or secretion of at least one toxin, and (ii) the mutant produces less of the toxin than the parentcell when cultured under the same conditions; and (b) isolating the polypeptide from the culture medium. The present invention relates to novel host cells and to methods of producing proteins. More specifically the invention relates to a host cell useful for the expression of heterologous proteins, in which the host cell has been genetically modified in order to express significantly reduced levels of a metalloprotease and an alkaline protease.
