The invention describes a method for the isolation of components from samples, particularly large molecular weight DNA from biological samples. The method involves the application of controlled oscillatory mechanical energy to the sample for short periods of time of about 5 to 60 seconds to lyse the sample and release the component(s) from the sample, followed by standard isolation methods. In preferred embodiments, the method includes the use of a spherical particle for applying the mechanical energy.

The present invention contemplates a system for rapidly isolating nucleic acids. The system comprises an insoluble silica matrix and a buffered aqueous salt solution containing salt at a concentration of at least 3 molar and a buffering agent at a concentration sufficient to provide a buffering capacity corresponding to that which either tris(hydroxymethyl)aminomethane or phosphate ion at a concentration of 0.1 to 1 molar would provide in the solution. In a plastic 'miniprep' device for separating liquid samples including (i) a first, sample, cylindrical container having an inlet and an outlet opening between which a separation layer is arranged, the outlet opening typically being connected to and enclosed by an outlet spout, fitting within a cylindrical bore of (ii) a second, collecting, container receiving the separated liquid discharged from the outlet spout, the cylindrical first container is (a) of lesser diameter than is the bore, and (b) possessed of longitudinal exterior rib hat hold it eccentric within the bore. Multiple, typically 5+, trays of an identical special configuration are vertically stacked one atop the next, end-to-end reversed. Each tray has an overflow feature, normally in the form of either (i) a portion of a side that is reduced in height so as to form a lip, (ii) a corner hole protected by a removable baffle of selectable height, or (iii) a threaded standpipe that may be screwed to variable height.
