A method for determining if a tissue is ganglionic is provided. The method comprises a) generating at least one hyperspectral Raman image of a tissue from a region of interest in a tissue suspected to contain ganglion cells that was optionally identified by a method comprising autofluorescence (AF) and Second Harmonic generation (SHG) imaging of the tissue; and b) analyzing any of the images for one or more of the following: i) optical excitation; ii) chemical information or emission spectra; or iii) AF, SHG, and/or Raman signatures, wherein the analysis provides indicators that the region of interest is either ganglionic or non-ganglionic. A system for analysis of in vivo tissue or ex vivo tissue samples including a multiphoton autofluorescence microscope, a Second Harmonic Generation microscope, and a hyperspectral Raman microscope in operative communication is also provided.

Label-free spectral pathology for in vivo diagnosis

Raman spectra of cells, such as normal human T- and B-cells from peripheral blood or human tonsil and the corresponding transformed cells are obtained by optically trapping the cells and obtaining their Raman spectra. The trapped cells can be subjected to one, two, or more different excitation wavelengths, and each wavelength of the corresponding Raman spectra can be stored in a separate channel. In preferred embodiments, two spectra are subtracted from each other to give a difference spectrum and each channel is analyzed independently to characterize the trapped cell. Alternatively, the Raman spectrum can be subjected to Principal Component Analysis (PCA) in order to characterize the trapped cell. The trapped cell thus classified can be sorted, or further manipulated.

Single-cell raman spectroscopy for the non-destructive, non-invasive analysis of cells and cellular components

Methods and systems for reconstructing individual spectra acquired from laser interrogation spots in a 2D array illuminating a particle are described. A particle is positioned in a 2D array that includes multiple laser interrogation spots. The laser interrogation spots of the particle are detected in the 2D array using a spectrometer. Multifocal spectral patterns are generated based on the laser interrogation spots, and an individual spectrum for each laser interrogation spot is reconstructed based on the plurality of multifocal spectral patterns.

Parallel acquisition of spectral signals from a 2-D laser beam array

Methods are disclosed for isolation of a stem-cell derived differentiated cell, which method entails illuminating an incident light onto a plurality of stem-cell derived cells that comprise at least an undifferentiated cell and a differentiated cell possessing a noncentrosymmetric structure, wherein the differentiated cell generates second-harmonic light from the incident light; and isolating the differentiated cell identified by the second-harmonic light. Devices for carrying out the methods are also provided.

Label-free identification of stem cell-differentiated cells

A double-pass fiber-optic based spectroscopic gas sensor delivers Raman excitation light and infrared light to a hollow structure, such as a hollow fiber waveguide, that contains a gas sample of interest. A retro-reflector is placed at the end of this hollow structure to send the light back through the waveguide where the light is detected at the same end as the light source. This double pass retro reflector design increases the interaction path length of the light and the gas sample, and also reduces the form factor of the hollow structure.

Combined raman and IR fiber-based sensor for gas detection

Several methods have been found to make p-type gallium nitride. P-type gallium nitride has long been sought for electronic devices. N-type gallium nitride is readily available. Discovery of p-type gallium nitride and the methods for making it will enable its use in ultraviolet and blue light-emitting diodes and lasers. pGaN will further enable blue photocathode elements to be made. Molecular beam epitaxy on substrates held at the proper temperatures, assisted by a nitrogen beam of the proper energy produced several types of p-type GaN with hole concentrations of about 5.times.10.sup.11 /cm.sup.3 and hole mobilities of about 500 cm.sup.2 /V-sec, measured at 250.degree. K. P-type GaN can be formed of unintentionally-doped material or can be doped with magnesium by diffusion, ion implantation, or co-evaporation. When applicable, the nitrogen can be substituted with other group III elements such as Al.
