The present invention is drawn to methods of preparing a bioactive agent-containing emulsion for delivery to a biological system. This method can comprise the step of jetting a bioactive agent and a first fluid medium from a fluid-jet pen into a second fluid medium to form a bioactive agent-containing emulsion, wherein the second fluid comprises a continuous phase of the emulsion. Alternatively, a method of preparing a bioactive agent-containing liposome can comprise jetting a lipid-containing composition and a bioactive agent from a fluid-jet pen into a medium to form a bioactive agent-containing liposome carried by the medium. The present invention is also drawn to fluid-jet pens and systems configured for making liposome- and emulsion-containing biological agents.

Fluid-jet pens configured for making modulated release bioactive agents

A method of making microcapsules including activating a fluid ejector at a frequency greater than 10 kilohertz where each activation of the fluid ejector generates essentially a drop, and the fluid ejector is fluidically coupled to a first fluid including a core component. The method further includes ejecting the drop of the first fluid into a second fluid, the drop having a volume. In addition, the method includes generating a microcapsule, that includes the core component, in the second fluid for each drop of the first fluid ejected.

Method of making microcapsules utilizing a fluid ejector

An expanding tablet is described comprising a drug release controlling membrane material. After swallowing, the tablet hydrates and expands such that the membrane ruptures to directly expose some surfaces of the core tablet to hydrating and eroding liquids, thus generating in situ a tablet which is platform supported on non-exposed surfaces, and which releases active ingredient in approximately zero order fashion. More particularly, the dosage form is adapted for controlled release of various pharmaceuticals. A working embodiment of the tablet was a spray-coated tablet comprising a core having greater than 25% of an expandable material which expands upon exposure to an aqueous environment and at least one active ingredient, e.g., glipizide, and an outer rupturable coating surrounding the core comprising a rate release modifying membrane and a water-soluble modifier. A method for administering an active ingredient also is described. The method comprises (1) providing a tablet according to the invention, and (2) administering the tablet to a patient.

Coated, platform-generating tablet

Active agents comprising coated pellets which self seal if they are damaged are described. More specifically, an active agent, or bead coated with an active agent, is coated with a rate-release controlling polymer and a hydrophilic gel-forming material which forms a gel upon hydration. If the bead is compressed into a solid compact which damages the polymer coating, the hydrophilic substance gels upon exposure to an aqueous environment. The gel provides sufficient sealing of the damaged area in the polymer so that a useful control of drug release is retained in spite of the damage to the polymer. The pellets of the invention exhibit improved flow and compactability. The compacts can be formulated to disintegrate in the gastrointestinal tract, and also may result in either controlled release or immediate release of the active agent.

Methods of treating circadian rhythm phase disorders

A peptide, produced by propionibacteria, has a molecular weight of between 300 and 1200 daltons and is inhibitory to gram-negative bacteria. The peptide can be produced by purification of a propionibacteria metabolite mixture, by chemical synthesis, or by a host transformed with a recombinant vector, and is useful in preventing and treating bacterial infections.

A metabolite material of propionibacteria is added to a food product to inhibit the growth of yeast. The metabolite material is produced by growing propionibacteria cells in a liquid growth medium to produce a mixture containing the metabolite material. The mixture can be concentrated and added to a food product as a concentrated liquid or powder. The metabolite material added to a food product may contain viable cells of propionibacteria.

Propionibacteria metabolites inhibit spoilage yeast in foods

A metabolite material of propionibacteria, having a metabolite of molecular weight greater than 300, is added to a food product to inhibit the growth of gram negative psychotropic bacteria, yeast, mold, gram positive bacteria, or Listeria. The metabolite material may contain less than 0.02% propionic acid such that there is insufficient propionic acid per se to inhibit microbial growth. The metabolite material is produced by growing propionibacteria cells in a liquid growth medium to produce a mixture containing the metabolite material. The mixture can be concentrated and added to a food product as a concentrated liquid or powder. The metabolite material added to a food product may contain viable cells of propionibacteria.

This invention relates to the preservation of high moisture animal feeds which utilize a fermentation (either natural or induced) to improve the quality of the animal food. More specifically, this invention utilizes dimethyl fumarate (DMF) in plant feeds in combination with fermentation microorganisms to produce an animal feed which is high in nutrient value and low in spoilage microorganisms.

The invention comprises a novel starter medium for the commercial propagation of acid producing bacteria, such as those used in food fermentation processes. The compositions are unique in that they contain a highly effective buffering ingredient which is a sodium, potassium, or ammonium salt or double salt of a linear aliphatic dibasic acid having from three to seven carbon atoms. The salts are present in an amount sufficient to maintain the growth medium at pH levels of about 5.0 or above during the time in which the bacteria are multiplying in the culture medium. Disodium or diammonium succinate, glutarate, or adipate are materials which have been found to be particularly effective. These may be used in combination with nutrients such as whey, whey permeate, nonfat dried milk, yeast extract, and diammonium phosphate. The addition of trace quantities of certain metals promotes the growth and activity of the acid producing bacteria. Small quantities of ferrous, manganous, or manganese ions are particularly useful. A combination of iron and manganese with the other ingredients in the media produced results better than either of these materials standing by itself. In a commercial test, the cheese produced using an inoculant based on one of the present formulations was of excellent quality. Most of the formulations containing the bibasic acid salts appear to be highly resistant to bacteriophage infection.

Method and buffered bulk starter media for propagation of useful bacteria

An improved powdered bacterial growth medium composition adapted to be admixed with water is described. The powdered growth medium includes an alkaline earth metal cation in a compound A admixed with a compound B containing an anion which reacts with the alkaline earth metal cation in compound A in an aqueous growth medium to form an essentially water insoluble salt or base, including the alkaline earth metal cation and the anion, which is acid neutralizing. The water insoluble salt is thus formed in situ in the aqueous growth medium when compounds A and B are added to the aqueous solution. Also described is an improved method for growing acid producing bacteria in an aqueous growth medium by forming the insoluble salt or base. The resulting growth medium is particularly adapted for neutralizing acids generated during growth of lactic acid producing bacteria which are grown for use in various food fermentations.

Bacterial growth medium and method of use

Methods for the management of bronchopulmonary insufficiency and other conditions are disclosed. The methods involve administration of an N-7-substituted derivative of theophylline selected from the group consisting of dyphylline, etophylline, proxyphylline and mixtures thereof. The derivative can be administered intravenously, intramuscularly, or in the lungs, concurrently with some form of theophylline or administered alone to a patient whose body has a known or an unknown theophylline content. Theophylline and the derivative can be administered in the same dosage form or in separate dosage forms. They can have the same route of administration or different routes of administration; for example, theophylline can be given orally, while the derivative is given as a zero-order intravenous infusion. A high total plasma concentration of methyl xanthine is achieved safely by using a combination of theophylline and one or more of the derivatives. Rapid injection of a loading dose of the derivative is also disclosed.
