Systems and methods for delivering a therapeutic agent to target tissue using a plurality of drug delivery platforms each having a polymer scaffold and the therapeutic agent contained within the polymer scaffold. Each drug delivery platform of the plurality of drug delivery platforms is implanted into the target tissue, allowing the therapeutic agent to elute from each drug delivery platform of the plurality of drug delivery platforms for a target period of time, delivering a therapeutic dose of the therapeutic agent for the target period of time. Each drug delivery platform of the plurality of drug delivery platforms is allowed to biodegrade within the target tissue.

Submucosal bioresorbable drug eluting platform

Biodegradable scaffold for wound closure including a central rod and upper and lower plates that each at least partially circumscribe the central rod. Each of the upper and lower plates is attached to the central rod and includes an upper section and a lower section that are connected by a helical middle section. The upper section and lower section of the upper plate respectively include first and second bottom surfaces and the upper section and lower section of the lower plate respectively include first and second top surfaces that are respectively spaced apart from and face the first and second bottom surfaces. The first and second bottom surfaces and first and second top surfaces may lie in different planes that are each perpendicular to a central longitudinal axis of the central rod.

Implantable tissue scaffold

Described here are devices, systems, and methods for treating conditions or diseases of the nose, ear, or throat with an expandable device having a drug coating. The expandable devices may be delivered to a body cavity in a low-profile configuration and expanded to contact surrounding tissue. The expandable devices may deliver or release the drug coating to the tissue. Multiple expansions of a single device may be employed during treatment. Various coating excipients and manufacturing parameters for the expandable devices may also be adjusted to enhance or slow transfer of the drug coating and/or release of the drug to the target tissue site. The drug transferred to the tissue may act as an in situ depot that enables maintenance of a therapeutic level of locally delivered drug for a desired time period after removal of the expandable devices.

Drug-coated balloon

A biodegradable scaffold for wound closure including a central rod and two spaced-apart plates. The central rod may have an engagement block on top for stabilizing an implanting tool. The two spaced-apart plates include an upper plate connected to the central rod and a lower plate connected to the central rod. Each of the upper and lower plates may include perforations which may be useful for detachably connecting an implanting tool and which encourage ingrowth of the tissue. The biodegradable scaffold allows tension free anatomic alignment of tissue within the plates and facilitates wound healing. The upper and lower plate may have a helical portion, in which each plate may extend around the central rod. The helical portion serves to draw or pull tissue around the wound defect into the scaffold in the course of deploying the scaffold.

Described here are systems and methods for delivering an active agent to target tissues of the ear, nose, or throat using an expandable member having drug crystals layered thereon, and methods for manufacturing such systems. The expandable member can be delivered to the target tissues in a low-profile configuration and expanded to contact and/or dilate surrounding tissue. Expansion of the expandable member transfers the drug crystals to the target tissues, which then act as an in situ depot that enables maintenance of a therapeutic level of an active agent for a desired time period after removal of the expandable member. Multiple expansions of a single expandable member can be employed during treatment. For example, the systems and methods can be useful when it is desired to treat multiple paranasal sinuses with a single expandable member.

Expandable member systems and methods for drug delivery

A biodegradable device for maintaining the alignment of the edges of a trocar defect, consisting of two bases coupled and offset by a connector. The first base to be positioned below the defect and a second base above. The first base has a threaded hole from its upper surface but not through the lower surface. The connector attached to the bases such that there is a hole aligned with the threaded hole in the first base allowing a device to mate with the threads in the first base. The second base has a hole aligned with the hole in the connector and wide enough to allow a device to mate with the threads in the first base. The device is arranged so the distance between the lower surface of the second base and upper surface of the first base holds the fascia around the trocar defect.

Implantable scaffold and method

A method for maintaining the alignment of the edges of a trocar defect by inserting into a defect a device for compressing the tissue such that part of the device is above while the other is below the defect, then pulling back on the device so that the part below the defect is up against the defect, then while holding the device below the defect in place, push down on the part above the wound so that it compresses the tissue. Finally, release the device from the insertion tool.

Biodegradable apparatus and method for closure of trocar defects

An ophthalmic shunt includes a seal or septum through which multiple injections of the therapeutic agent can be made as needed, thereby avoiding repeated injury to the sclera. The therapeutic agent is temporarily carried in a tube connected to the septum. The therapeutic agent release may be rapid or sustained over a period of time.

Trans-sclera portal for delivery of therapeutic agents

A polymeric stent can be implanted for treatment of the Eustachian tube. The stent can be designed to have length-dependent radial strength to allow it to stay within the Eustachian tube and to allow normal closing and opening of the Eustachian tube. A balloon can be used to implant the stent, and the balloon can be coated with a therapeutic agent. A coated balloon can also be used to transfer therapeutic agents to the sinus cavity during a balloon sinus dilation procedure.

Devices and methods for treatment of the Eustachian tube and sinus cavity

Formulations and methods are disclosed which provide controlled, sustained release of a biologic therapeutic to a space within the body. More specifically, formulations comprising a plurality of hydrophilic polymer strands, and methods of forming and administering such formulations, are disclosed. In some embodiments, the formulations exhibit a burst release, an initial release, a triphasic release, and release over thirty to ninety days of the biologic therapeutic. In some embodiments, the formulations exhibit reversible precipitation of the biologic therapeutic into precipitates having a diameter of about 50 nm to about 10 &#x3bc;m.
