The present disclosure pertains to an apparatus and methods of an imaging device for obtaining images from the walls of luminal organs or a surgical cavity. The device is capable of passage through luminal organs or introduction into surgical cavities, and obtains images by rapidly scanning a focused light beam on the tissue to be imaged and receiving light from the tissue. The device comprises at least one mechanism for projecting a 2-dimensional (2-D) optical pattern, such as a 2-D scanner (508) or a 2-D optical array forming a closed loop scan pattern (506). The device comprises at least one other beam scanning mechanism, such as a rotary or angular scanner (510) such that cycloid scan pattern is formed on the surface to be scanned, and also has embodiments of focusing optics at different regimes of numerical aperture. The disclosure also describes methods for accurate image reconstruction with embodiments of hardware control and post-acquisition processing.

Scanning optical imaging device

A computer-implemented method of imaging an object, and an optical coherent tomography (OCT) imaging system implementing same. The method comprises acquiring a three-dimensional optical coherence tomography (OCT) data set representing an object, wherein the OCT data set includes at least a first and a second three-dimensional data subsets, each element of the OCT data set having a respective sampling period, wherein at least a first element of the first data subset represents a point in space that is not represented by any element of the second subset, and at least one element of the second subset has a sampling period different from the sampling period of the first element of the first subset; processing at least the first and the second data subsets according to at least one imaging modality, thereby generating at least a first and a second processed data subsets, each processed data subset representing the object; and generating a composite image representing the object based on at least the first and the second processed data subsets.

An apparatus and method for real-time optical imaging of a tissue specimen. The apparatus comprises a primary imaging system configured to use an illumination source to acquire images of a tissue specimen through one or more spectrally separated channels, and configured to perform optical depth sectioning; an auxiliary imaging system, configured to acquire an auxiliary image of the tissue specimen; a specimen holder having a transparent window therewithin, window, the specimen holder comprising one or more position sensors, wherein the specimen holder is configured to be translatable in the specimen plane; a user input device configured to accept user input, wherein the specimen holder is configured to translate in response to the user input in real-time; a processing unit configured to execute a sequence of instructions on the sequence of images acquired by the primary imaging system, the auxiliary image, and at least one specimen holder position to generate a composite representation of the tissue specimen that includes a representation of cell nuclei in the specimen; and a display device configured to display the composite representation of the tissue specimen in real-time.

Images of an object, such as OCT scans of a human eye, can include distortions and data gaps due to relative motion of the object and the image acquisition device. Methods and systems for correction of such distortions and data gaps are described herein. Motion-corrected data is arrived at by applying three-dimensional transforms to input three-dimensional data sets that represent at least partially overlapping regions of the imaged object. The three dimensional transforms are computed based on an objective function that accounts for similarity between the transformed three-dimensional data sets and the estimated motion of the object relative to an imaging instrument. Methods and systems described herein advantageously eliminate the need for postulated assumptions and reliance on landmarks and are capable of filling data gaps, thereby producing high quality, undistorted images of objects subject to movement during imaging. Multiple motion-corrected data sets can be merged or combined to produce a data set with improved image quality.

Method and apparatus for motion correction and image enhancement for optical coherence tomography

A control system for improving and stabilizing Fourier domain mode locking (FDML) operation. The control system may also provide regulation of FDML operational parameters such as filter tuning, laser gain, polarization, polarization chromaticity, elliptical polarization retardance, and/or dispersion. In one aspect, the invention relates to a method of acquiring optical coherence tomographic data from a sample. The method includes the steps of scanning a first location on the sample to obtain a first set of optical coherence tomographic data, scanning a second location on the sample to obtain a second set of optical coherence tomographic data, and defining a fiducial position relative to a location on the sample using one of the two sets of optical coherence tomographic data. In one embodiment, the first set of optical coherence tomographic data is survey data. However, in another embodiment the first set of optical coherence tomographic data is sample measurement data.

In one aspect the invention relates to a frequency varying wave generator. The generator includes a gain element adapted to amplify a wave having a wavelength; a time varying tunable wavelength selective filter element in communication with the gain element, the tunable filter element adapted to selectively filter waves during a period T; and a feedback element in communication with the tunable filter element and the gain element, wherein the tunable wavelength selective filter element, the gain element and the feedback element define a circuit such that the roundtrip time for the wave to propagate through the circuit is substantially equal to a non-zero integer multiple of the period T.

A mirror system for use in generating a short duration laser pulse is disclosed. The system includes first and second double-chirped mirrors disposed along an optical path within a cavity, where the second double-chirped mirror includes an additional phase-shifting layer as compared to the first double-chirped mirror. The additional phase-shifting layer causes the mirror system during use to produce a laser pulse that is characterized by oscillations in group delay substantially reduced in amplitude in comparison to oscillations in group delay for a pulse produced by the same system without the additional phase-shifting layer.

A fiber optic needle probe for measuring or imaging the internal structure of a specimen includes a needle defining a bore, an optical fiber substantially positioned within the bore, and a beam director in optical communication with the optical fiber. At least a portion of the wall of the needle is capable of transmitting light. The beam director directs light from the optical fiber to an internal structure being imaged and receives light from the structure through a transparent portion of the wall. An actuating device causes motion of any, or all of, the needle, optical fiber, and beam director to scan the internal structure of the specimen. The fiber optic needle probe allows imaging inside a solid tissue or organ without intraluminal insertion. When used in conjunction with an OCT imaging system, the fiber optic needle probe enables tomographic imaging of the microstructure of internal organs and tissues which were previously impossible to image in a living subject.
