Disclosed herein are compositions comprising recombinant arenavirus monoclonal antibodies and antigen-binding fragments thereof, as well as therapeutic methods using the antibodies. In some embodiments, the antibodies provide pan-arenavirus protection against a number of arenavirus types and strains.

Arena virus monoclonal antibodies and uses

Disclosed herein are compositions comprising arenavirus monoclonal antibodies, as well as therapeutic, diagnostic, and preventative methods using the novel antibodies. Preventative methods include preparation of vaccines, as well as factors (e.g. small molecules, peptides) that inhibit Old World arenavirus infectivity, including LASV and LCMV. In some embodiments, the antibodies provide pan-arenavirus protection against a number of arenavirus types and strains. Diagnostic and therapeutic antibodies including neutralizing antibodies for the prevention and treatment of infection by LASV and other arenaviruses are also disclosed, as well as new tools and methods for the design, production, and use of arenavirus monoclonal antibodies, including expression in engineered bacterial- and mammalian-based systems.

Arenavirus monoclonal antibodies and uses

Methods and compositions are provided for the use of an envelope polypeptide or a functional variant thereof from a lentivirus that is not HIV-1 as a molecular scaffold for HIV-1 epitopes. The HIV-1 epitopes can be recognized by HIV-1 binding antibodies, HIV-1 neutralizing antibodies and/or CD4-induced antibodies. Thus, methods are provided for detecting HIV-1 binding antibodies in a subject infected with HTV-1. Further provided are methods to determine an epitope for an HIV-1 binding antibody; methods to assay for an HIV-1 binding antibody; methods to identify a soluble CD4 mimic; methods to neutralize an non-HIV-1 virus; diagnostic assays to monitor HIV disease in a subject or to monitor the subject's response to immunization by a HIV vaccine; and methods to alter the neutralization potential of an HIV-1 derived CD4-induced antibody. Chimeric polypeptides, chimeric polynucleotides, kits, cells and viruses are also provided.

The invention features immunoreagents which neutralize the Human Immunodeficiency Virus Type 1 (HIV-1) by binding to a novel conserved epitope of the HIV-1 gp120. These immunoreagents exhibit a broad neutralizing effect upon HIV attachment to host cells, and are therefore useful in the detection, prevention, amelioration and treatment of HIV disease, primarily AIDS (Acquired Immunodeficiency Syndrome) and ARC (AIDS Related Complex). A process for selectively immobilizing viral glycoproteins on lectin-coated surfaces for use in solid phase immunoassays is disclosed.
