The present invention provides a new fungal production system comprising a fungal host strain ofwherein the endogenous cellulase secretion is less than 20% of the endogenous cellulase secretion ofstrain UV 18-25. Preferably, also the secretion of endogenous protease, endogenous &#x3b2;-glucanase and endogenous cellobiohydrolase is less than 20% of the secretion ofstrain UV 18-25. Furthermore, fungal host strains are provided wherein several genes have been disrupted. According to another aspect of the invention a method for homologous and/or heterologous production of a pure protein with a purity of higher than 75%, comprising expressing a gene encoding said protein in the strains according to the invention have been described. Furthermore, a method for production of artificial protein mixes comprising expressing a gene encoding each of said proteins in a strain according to the invention have been disclosed. Finally a method for simplified screening of strains functionally expressing a desired enzyme by application of said strains have been provided.

Fungal production system

The present invention is in the area of enzymes for (hemi-)cellulose degradation and/or modification, more in particular the degradation and/or modification of xylan. The invention is based on a newly discovered enzymatic activity of a class of lytic polysaccharide monooxygenases (LPMOs), i.e. oxidative cleavage of xylan in addition to oxidative cleavage of cellulose. The present invention therefore relates to a method for degrading and/or modifying xylan in a xylan-comprising substrate, a method for preparing a product from a xylan-comprising substrate, a kit of parts, a liquid, paste or solid formulation, and a xylan-comprising composition, comprising said LPMO. The invention further relates to a use of said LPMO, said kit of parts, said formulation and/or said composition, in a method of the invention.

An enzyme system is described that is useful for preparing food and feed. One enzyme of that system is obtainable from Aspergillus. That enzyme has the following characteristics: a MW of from 29 kDa to 36 kDa as measured on a SDS-Phastgel (10-15%) or about 30 kDa; a pI value of about 3-4; ferulic acid esterase activity; a pH optimum of about 5; and a temperature optimum of from about 50 to about 60.degree. C.

An arabinoxylan-degrading enzyme from Aspergillus is provided. The enzyme is active in the degradation of water-insoluble solids obtained from maize. The enzyme can be used in the preparation of animal feed compositions, human food or in industrial processes.
