Compositions of matter comprising RNA silencing molecules capable of mediating cleavage of p21 mRNA are disclosed. A composition of matter comprising a DNAzyme molecule capable of mediating cleavage of p21 mRNA corresponding to SEQ ID NO: 1, wherein said DNAzyme molecule comprises a nucleic acid sequence at least 80% identical to the nucleic acid sequence set forth in any one of SEQ ID NOs: 23, 29, 33-38, 40, 42, 45-48, 53-60, 63-65, 69-74 or 78, is disclosed. Methods of eradicating senescent cells or cancer cells, as well as methods of treating senescence-associated diseases or disorders, cancer, and fibrotic diseases and disorders are also disclosed.
