In one aspect, the present invention is directed to a method for preventing or treating necrotic enteritis by administering a hyperimmunized egg product obtained from an egg-producing animal to an avian. The hyperimmunized egg product may contain an antibody specific to an antigen selected from the group consisting of&#x3b1;-toxin,elongation factor Tu (EF-Tu),necrotic enteritis B-like (NetB) toxin,Pyruvate: Ferredoxin oxidoreductase (PFO), andelongation factor 1-alpha.

Provided herein are immunogenic compositions containing recombinant proteins capable of presenting all, or antigenic portions of, theElongation Factor 1 alpha, or EF-1&#x3b1;, protein in the development of active immunity to, and control of, coccidiosis. Also provided are methodologies of using the immunogenic compositions for administration to poultry and other animals in the control of coccidiosis. In some instances, the EF-1&#x3b1; protein utilized in the immunogenic composition presented herein is molecularly manipulated or combined with adjuvants to increase effectiveness.

NK-2 synthetic peptide derived from cationic core region of porcine NK-lysin has an antimicrobial and antitumor polypeptide that is considered to play a pivotal role in innate defense immunity. To evaluate the antimicrobial properties of synthetic chicken NK-lysin peptides, we investigated cytolytic activity against apicomplexan parasites likesporozoites,tachyzoites andsporozoites. The chicken NK-2 (cNK-2) lytic peptide which corresponds to amino acid residues of porcine NK-2 peptide significantly disruptedandsporozoites andtachyzoites. In contrast, no bactericidal activity was observed onBMH71-18. The cNK-2 lytic peptide is novel antimicrobial agent which can be used to intervene and treat economically costly infections in the animal industry.

Antimicrobial activity of chicken NK-2 peptide against apicomplexan protozoa

This study reports a novel immunopotentiating effect of a lectin (FFrL) extracted from the mushroomon poultry cell-mediated immunity and poultry coccidiosis. We describe the extraction of FFrL, its in vitro mitogenic activity and in vivo protection against an oral challenge infection with. When tested on several cell types, crude FFrL agglutinated mouse erythrocytes and thymocytes and also various other cells including murine and human cell lines. However, crude FFrL did not agglutinate human A, B, AB, or O erythrocytes. FFrL showed a potent mitogenic activity on chicken splenic lymphocytes where at lower concentrations, crude FFrL exerted stronger mitogenic activity than Con A, a well-known potent mitogen for T lymphocytes. Further, FFrL significantly induced (P&#x3c;0.05) nitric oxide secretion in HD11 cells, an established macrophage cell line, and suppressed (P&#x3c;0.05) RP9 tumor cell growth, both, in a dose-dependent fashion. When injected into eighteen-day-old chicken embryos followed by an oralchallenge infection, FFrL-treatment significantly protected chickens against weight loss associated with coccidiosis (P&#x3c;0.05). Injecting embryos with FFrL also resulted in significant reduction in oocyst shedding as compared to the control saline-injected birds (P&#x3c;0.05). The results of this study demonstrate that FFrL is an effective growth-promoting and immunostimulating agent in poultry during coccidiosis.

This invention relates to a novel DNA encodingglyceroaldehyde-3-phosphate dehydrogenase (GAPDH) and recombinant GAPDH protein expressed using the DNA sequence. The present invention further provides the recombinant vector or virus containing this DNA. The chickens immunized by the recombinant virus or recombinant GAPDH protein described herein can be induced immunity against coccidiosis.

DNA encoding Eimeria glyceroaldehyde-3-phosphate dehydrogenase and uses thereof

A stable chicken hybridoma secreting a monoclonal antibody (mAb) that detects the conoid structure of ( ) sporozoites has been developed. The hybridoma is made by fusing a thymidine kinase (TK)-deficient chicken myeloma with spleen cells from chickens immunized with sporozoite antigen. The monoclonal antibody recognizes sporozoite proteins on the conoid of the anterior tip of sporozoites. The monoclonal antibody has been shown to inhibit the invasion of sporozoites into CD8 T cells in vitro thereby indicating its role in the recognition of host cells during the invasion process following infection with Eimeria parasites.

Chicken monoclonal antibodies specific for coccidial antigens involved in invasion of host lymphocytes

Seven murine hybridoma cell lines capable of secreting monoclonal antibodies that can identify subpopulations of chicken lymphocytes have been developed. Precursor cytotoxic and suppressor T lymphocytes, precursor helper lymphocytes, and mature T lymphocytes may be distinguished by use of these antibodies.

Monoclonal antibodies against chicken T-lymphocytes

Disclosed are DnA sequences which code for antigenic proteins, methods for identifying such DNA sequences, and antigens coded for by such DNA sequences. The first step of the method is to provide a multiplicity of DNA sequences. These sequences are then inserted into DNA expression vectors to form recombinant expression vectors. The expression vectors are inserted into suitable hosts to form transformants which express the DNA sequences. The transformants are then contacted with antibodies directed against Eimeria antigens to identify transformants containing DNA sequences which code for Eimeria antigens. These antigens are then produced from the DNA sequences identified as coding for the antigens. The antigens so produced are contacted with white blood cells which effect a cell-mediated immune response, which white blood cells are sensitized to an antigenic Eimeria protein, to thereby identify DNA sequences which code for antigens that induce a cell-mediated immune response to avian coccidiosis. The DNA sequences of the present invention comprise cloned genes or fragments thereof that code on expression for an antigenic protein that activates white blood cells which effect a cell-mediated immune response, which white blood cells are sensitized to an antigenic Eimeria protein.
