The present invention relates to a composition for promoting the efficiency of somatic cell replication, comprising endothelin as an active ingredient. When somatic cells are treated with the composition of the present invention, H3K9me3, which is suspected to be a factor for the lowering success rate of nuclear substitution, is inhibited, and H3K4me3, which has the opposite effect, is increased to increase the success rate of nuclear substitution. Accordingly, the composition of the present invention increases the efficiency of somatic cell replication, thereby increasing the efficiency of producing pluripotent stem cells derived from nuclear substitution, and ultimately, the composition is expected to contribute to the development of cell therapeutic agents.

Composition for promoting efficiency of somatic cell replication

Provided are a protein complex in which estrogen receptor 2 (-ERT2) is fused to CRISPR associated protein 9 (Cas9), and a recombinant vector carrying a gene coding the protein complex, wherein ERT2 is bonded to the N-terminus and C-terminus of nuclear localization sequence (NLS)-removed Cas9 and the complex has the advantage of translocating from the cytosol into the nucleus at a certain time point upon treatment with tamoxifen and modifying a specific DNA with the aid of guide RNA (gRNA), ultimately enabling a more elaborate DNA modification operation in a desired part at a desired time point.

Disclosed are a method for preparing induced pluripotent stem cells from endocardium-derived adult stem cells isolated from peripheral blood and a method for differentiating induced pluripotent stem cells into cardiovascular cells. The endocardium-derived adult stem cells are primary culture cells for preparing induced pluripotent stem cells, can be readily isolated and cultured only with a small amount of peripheral blood, have a high proliferation property so as to be storable without generic variation, and can rapidly ensure a cell number so as to be usable in cell therapy. The endocardium-derived adult stem cells have sternness, thereby having high preparation efficiency, and are derived from the endocardium so as to have the epigenetic memory of cardiovascular cells, thereby having an advantage of being able to be differentiated, after preparing induced pluripotent stem cells, into cardiovascular cells such as endothelial cells, smooth muscle cells and cardiomyocytes with a high efficiency.

The present invention relates to a method of preparing vascular endothelial cells by transforming (transdifferentiating) adult fibroblasts and a composition, which includes vascular endothelial cells prepared according to the method, for preventing and treating ischemic diseases, the method including a step of transducing adult fibroblasts with a gene. In particular, the present invention confirms that five factors, Foxo1, Er71, Klf2, Tal1, and Lmo2, induce transdifferentiation of adult fibroblasts into induced vascular endothelial cells. Furthermore, the present invention confirms that three factors, Er71, Klf2, and Tal1 induce transdifferentiation of human adult fibroblasts into induced vascular endothelial cells. The resultant induced endothelial cells enable lower limb salvaging by angiogenesis in lower limb ischemic animal models, showing that the induced endothelial cells can be effectively used for prevention or treatment of ischemic diseases.

Provided are a composition including a Kang AI 1 (KAI1) polypeptide or a gene encoding the same, for inhibiting angiogenesis, and a use thereof, and more specifically, a composition including a KAI1 polypeptide or a gene encoding the same, for inhibiting angiogenesis, and a pharmaceutical composition for preventing and treating cancer. Further, KAI1, which is expressed in a pericyte rather than in an endotheliocyte, can inhibit angiogenesis by functioning as a negative angiogenic regulator, and thus the subject matter can inhibit angiogenesis due to a known angiogenesis factor by various methods such as a method using a supernatant in a pericyte which forcefully increases KAI1 or in which KAI1 is forcefully increased, a method using a KAI1 protein, or a method for inhibiting a Src-Pkc pathway provoking the degradation of KAI1. Further, the subject matter can identify the severity of a cancer patient by identifying the expression of KAI1 in a pericyte.

The present invention relates to a method for increasing the stemness of human mesenchymal stem cells and, more particularly, to: a method for increasing the stemness of human mesenchymal stem cells by means of endothelin-1 treatment; the human mesenchymal stem cells having increased stemness by using the method; and a composition for increasing the stemness of human mesenchymal stem cells, containing endothelin-1 as an active ingredient. In the present invention, it is confirmed that the expression of a stemness marker is increased and that a stem cell characteristic is improved such as the length of telomeres being extended, by treating human mesenchymal stem cells with endothelin-1, and thus cellular life span is extended, aging is inhibited, and the growth and viability of cells are increased, thereby enabling mass culturing of human mesenchymal stem cells such that human mesenchymal stem cells are expected to be used effectively in cell therapy or regenerative medicine.

The present application discloses a method for producing peripheral blood stem cells with improved angiogenic properties through EPO or APS priming, and a use thereof.
