Immunoreactive heterochain antibodies are described. The heterochain antibodies are made up of light chain and heavy chain variable regions derived from different antibodies. The heterochain antibodies can exhibit antigen binding properties which are different from the parent antibodies from which they are derived. Methods of producing the heterochain antibodies and methods of their use in diagnostic and therapy are also disclosed.

Immunoreactive heterochain antibodies

The monoclonal antibodies are specific for a determinant found on hepatitis B surface antigen, and show high affinity for this determinant. Hybridomally produced monoclonal IgM antibodies having high affinity are useful for the immunoassay and purification of viral antigens.

A simultaneous sandwich immunoassay employing high-affinity monoclonal antibodies is disclosed. This simultaneous sandwich assembly has surprising sensitivity compared to forward and reverse sandwich assays for the detection of multi-determinant antigens such as hepatitis B surface antigen.

Immobilization of active anti-cortisol antibodies onto negatively charged solid phase surfaces results in a shift of the antibody pH optimum to an acidic range thereby permitting effective yet minimal use of deblocking agents and an extraction-free immunoassay for cortisol.
