The present disclosure provides novel methods and compositions for the maturation and regeneration of plantlets from microspore-derived embryos. The compositions provided include a liquid maturation composition comprising a first plant hormone and about 430 mM to about 880 mM of a monosaccharide solute, a disaccharide solute, or a polysaccharide solute. The methods provided include the steps of contacting a microspore-derived embryo with a liquid maturation medium for a time period sufficient to produce a matured microspore-derived embryo, transferring the matured microspore-derived embryo to a substrate, and regenerating the plantlet from the matured microspore-derived embryo.

Methods and compositions for improved plant regeneration from microspore-derived embryos

The invention provides novel methods for liquid-mediated delivery of pollen grains to enclosed stigmas in recipient female flowers. For example, methods for liquid-mediated pollination are provided. The methods provided include collecting pollen from a donor plant, suspending the collected pollen in a liquid solution, and introducing said solution to an enclosed stigma of a recipient flower bud on a recipient plant, thereby pollinating the flower with the pollen from the donor plant.

Provided are methods for increasing the efficiency of creating doubled haploid plants by increasing the number of chances of forming a double haploid seed through treatment of a monocot plant with a plant growth regulator. In certain embodiments, maize plants are produced that comprise multiple co-dominant ears. Also provided are plants comprising the potential to generate increased numbers of doubled haploid offspring.

A plant support for centrifuging plants comprises at least first and second portions that are configurable in a closed configuration that defines an internal cavity adapted to radially support plants in a manner at least partially preventing the plants from deforming or changing orientation during centrifugation. Provided herein are delivery methods of a plant treatment agent to a tissue of a plant, wherein the tissue is contacted with a plant treatment agent and delivery is aided by a centrifugal force applied to the plant. Disclosed herein is the discovery that drastic improvements in treatment delivery occur when plants are centrifuged after being contacted, submerged, unsubmerged, soaked, etc. in a solution containing a plant treatment agent. In one embodiment, haploid plants contacted with a colchicine solution are centrifuged to reveal improved chromosomal doubling rates as compared to control plants.
