The present invention provides a method for efficiently manufacturing a non-enveloped virus with high purity without laborious operation by cultivating cells having the ability to produce a non-enveloped virus and bringing the cells and an acidic solution into contact with each other. A non-enveloped virus vector manufactured by the method of the present invention and a composition having the non-enveloped viral vector as an active ingredient are very useful as gene transfer methods in the fields of basic research and clinical application gene therapy.

Method for manufacturing non-enveloped virus

The present invention provides a method for producing a virus vector, which involves a step of culturing a cell capable of producing the virus vector in a culture medium containing a retinoic acid compound, a histone deacetylase-inhibiting substance and a substance capable of forming a chelate; and a culture medium for use in the production of a virus vector, which is characterized by containing a retinoic acid compound, a histone deacetylase-inhibiting substance and a substance capable of forming a chelate as active ingredients.

Method for producing viral vector

The present invention provides a method for producing a virus vector, which comprises a step wherein cells that are capable of producing a virus vector are cultured in a culture medium that contains, as active components, a retinoic acid and a histone deacetylase inhibiting substance; and a culture medium for the production of a virus vector, which is characterized by containing, as active components, a retinoic acid and a histone deacetylase inhibiting substance.

Method for producing virus vector

A method for obtaining a virus vector free of a serum, and a serum-free medium for cultivation of a virus producer cell which can be used for the method are provided. By cultivating a virus producer cell using a serum-free medium containing serum albumin, it is possible to cultivate the cell in a state equivalent to that in a serum-containing medium to produce a virus vector at a sufficient titer. The virus vector prepared from the virus producer cell cultivated in the medium exhibits gene transfer efficiency comparable to that of a conventional vector. The medium is useful for gene therapy and studies thereof.

Serum-free medium for producing retroviruses

Improved methods for transferring a gene into target cells by using a retrovirus, wherein the gene transfer efficiency is improved and the target cells are efficiently transformed by binding the retrovirus to a functional substance which is immobilized on as carrier and having an activity of binding to retroviruses followed by washing; using an antibody capable of specifically recognizing cells, laminin or mannose-rich type sugar chain as a substance having an activity of binding to the target cells; pre-treating the target cells so as to inactivate transferrin receptor, or introducing a new functional group into the functional substance.

Gene transfer methods

An isolated DNA is provided coding for the variable region of an anti-human influenza A type virus antibody having the following characteristics: (a) specifically binds to the stem region of haemaggulutinin molecule of H1N1 and H2N2 subtypes of human influenza A type virus but does not specifically bind to the stem region of haemaggulutinin molecule of H3N2 subtype; and (b) has a neutralization activity for the H1N1 and H2N2 subtypes of human influenza A type virus but no neutralization activity for the H3N2 subtype.
