The invention relates to delivery systems that allow for the pulsatile release of a substance, such as a drug, in response to a change in pH. More specifically, it relates to drug administration to the GI tract, in particular to site-specific intestinal drug delivery via the oral route. Provided is a pH-controlled pulsatile release system (PPRS) comprising a core surrounded by a coating layer, wherein said core comprises an active substance and wherein said coating layer comprises a pH-sensitive coating material wherein a swellable agent is embedded. Said swellable agent is capable of taking up at least 1.1 times, preferably at least 5 times, more preferably at least 10 times its weight in water. Also provided is a pharmaceutical composition comprising a PPRS, in particular a colon-specific PPRS.

PH-controlled pulsatile delivery system, methods for preparation and use thereof

The invention relates to a time controlled, immediate release drug delivery system for oral administration of a first active ingredient to a subject in need thereof. The invention additionally relates to a dual drug delivery device, comprising the time controlled, immediate release drug delivery system according to the invention, further comprising a second coating comprising a second active ingredient.

The present invention relates to compositions and methods for the prophylaxis or treatment of deficiencies in essential amino acid absorption and metabolism and/or of a pathology or symptom associated there with. In particular the invention concerns the treatment and/or prevention of ADHD, ADD and autism spectrum disorders. The present inventors have developed a method for prophylaxis or treatment of such symptoms and/or pathologies associated with a deficiency in essential amino acid absorption and/or metabolism, which method, stated generally, relies on the administration of nicotinamide, typically in a long-acting formulation so as to overcome the deficiencies of existing formulations, which have proven unsuitable for effective treatment.

Nicotinamide compositions and the therapeutic use thereof

A breath actuated dry powder inhaler with a single air circulation chamber for de-agglomeration of entrained powdered medicament using the energy of the inspiratory air stream. The chamber has a substantially polygonal sidewall, a plurality of air supply channels entering the chamber substantially tangentially to its sidewall. A powder channel extends through a powder dose supply region of the inhaler tangentially into the chamber. An air outlet axially extends from a discharge and connects to a discharge channel that extends to a mouthpiece. The polygonal sidewall comprises at least six straight line segments, each straight line segments being spaced at the same first distance from an adjacent one forming the plurality of air supply channels. The air supply channels have the same width. The powder channel is defined by two straight line segments which are spaced from each other at a second distance which is larger than the first distance.

Breath actuated dry powder inhaler

A breath actuated dry powder inhaler, comprising a substantially disc shaped air circulation chamber for de-agglomeration of entrained powdered medicament using the energy of the inspiratory air stream. The chamber has a substantially circular or polygonal sidewall extending about a central axis between top and bottom walls of the chamber so that the height of the chamber is smaller than its diameter. A plurality of air supply channels disposed about the circumference of the chamber, which channels extend from joint or separate air inlets and which channels enter the chamber substantially tangentially to its sidewall. At least one of the supply channels extends through a powder dose supply region of the inhaler. The chamber further comprises an air outlet axially extending from a discharge opening in the centre of the top or bottom wall of the chamber and connects to a discharge channel that extends to a mouthpiece. The inhaler comprises at least one further air circulation chamber for de-agglomeration of entrained powdered medicament, the chambers being connected to the mouthpiece in parallel.

Dry powder inhaler and method for pulmonary inhalation of dry powder

The invention relates to a powder formulation for administration by inhalation comprising an active substance and a pharmaceutically acceptable excipient, which composition has the form of a physical mixture and comprises from 5 to 25 wt. % of the excipient, and wherein the active substance has a particle size distribution of from 0.5 to 10 &#x3bc;m, and wherein the excipient has a particle size distribution of from 15 to 500 &#x3bc;m.

Powder formulation

An antibacterial formulation suitable for injection into animals containing approximately 2-10% w/v difloxacin HCl, L-arginine base, propylene glycol, ethanol and/or benzyl alcohol, and water. The formulation is a solution having a pH of form 9 to 10. The formulation produces little or no tissue damage or irritation at the injection site.

The present invention relates to an auxiliary substance for an active substance, such as a pharmacon. The auxiliary substance has a stabilizing action. The auxiliary substance further has a positive influence on the bioavailability of the active substance with which the auxiliary substance can be incorporated into a pharmaceutical preparation. The auxiliary substance is based on a fructan having a number-average degree of polymerization of at least 6 and is used in the form of a sugar glass.

Stabilizer for pharmacons

A disperser for dry powders which can be used with different dose systems, dose weights ranging from 2 to 25 mg and different types of powder formulation. In one embodiment, the disperser acts both as a de-agglomeration (disintegration; aerosolization) means and as an air classifier for especially adhesive mixtures. Only fine drug particles are emitted whereas the larger agglomerates and carrier crystals are retained by the disperser. Another embodiment enables time controlled release of carrier crystals in these mixtures. Yet another embodiment has optimized performance with spherical pellets, containing no carrier crystals. Other possible embodiments of the invention make it possible to control the total inhaler resistance and the powder deposition in the upper respiratory tract by means of the addition of a so-called sheath flow of clean air. Modifications also enable carrier retainment in the mouthpiece and elimination of the tangential flow component of the discharge cloud.

Powder formulation disintegrating system and method for dry powder inhalers

The invention relates to a novel filler-binder for tablets, which shows a significantly reduced lubricant sensitivity when compared to prior art filler-binders. The invention further relates to a process of making said filler-binder and to the tablet obtainable by said process.
