Protease variants and uses thereof

Disclosed are compositions and methods relating to variant maltohexaose-forming alpha-amylases. The variant alpha-amylases are useful, for example, for starch liquefaction and saccharification, for cleaning starchy stains in laundry, dishwashing, and other applications, for textile processing (e.g., desizing), in animal feed for improving digestibility, and for baking and brewing.

The present disclosure relates to serine proteases cloned from, and variants thereof. Compositions containing the serine proteases are suitable for use in cleaning fabrics and hard surfaces, as well as in a variety of industrial applications.

Automatic dishwashing detergent composition

The present invention provides methods and compositions for the production of mature proteases in bacterial host cells. The compositions include polynucleotides encoding serine protease sequences with modified or heterologous propeptide regions; polypeptides comprising serine proteases with modified or heterologous propeptide regions; expression cassettes, DNA constructs, vectors, and chromosomes comprising such polynucleotides; and bacterial host cells comprising such polynucleotides. The methods include methods for enhancing the production of mature proteases in bacterial host cells (e.g.sp. host cells). The produced proteases find use in the industrial production of enzymes, suitable for use in various industries, including but not limited to the cleaning, animal feed and textile processing industry.

A method of oxidizing a saccharide, by contacting the saccharide with an alcohol dehydrogenase (ADH) enzyme selected from a quinone redox cofactor-dependent ADH, a nicotinamide adenine dinucleotide (NAD) redox cofactor-dependent ADH, a nicotinamide adenine dinucleotide phosphate (NADP) redox cofactor-dependent ADH, and any combination thereof is described. An oxidized saccharide obtainable by the method and products, in particular food products and paper products, containing the oxidized saccharide, are also described.

Oxidation process

An amino acid sequence is described. The amino acid may comprise a signal sequence that is SEQ ID NO. 1a (or a variant or homologue or derivative or fragment thereof) that is expressed as a fusion protein that comprises a protein of interest. The signal sequence directs secretion of the protein of interest. The protein of interest may be a heterologous protein. Secretion of the fusion protein aids purification.

We disclose a PS4 variant polypeptide derivable from a parent polypeptide, the parent polypeptide having non-maltogenic exoamylase activity, which PS4 variant polypeptide comprises one or more of the following substitutions: G69P, A141P, G223A, A268P, G313P, S399P and G400P, with reference to the position numbering of a Pseudomonas saccharophilia exoamylase sequence shown as SEQ ID NO: 1. Such PS4 variant polypeptides may be used as exo-amylases, particularly as non-maltogenic exoamylases. Combinations of such PS4 variant polypeptides together with Novamyl are disclosed.
