There is provided a device for partitioning a liquefied sample into discrete volumes. The device includes a bottom member; a top member disposed adjacent the bottom member; and at least one channel member disposed between the top and bottom members. The at least one channel member is at least partially defined by the top and bottom members and has first and second end portions. The first end portion of the at least one channel has an opening to receive liquid and the second end portion of the at least one channel has a reaction compartment and a vent opening. Accordingly, when the liquefied sample is introduced to the first end portion, capillary action assists in causing the liquefied sample to travel from the first end portion to the second end portion and at least a portion of the liquefied sample is caused to remain in the reaction compartment.

Apparatus and method for liquid sample testing

The invention provides devices and methods for rapid quantification of microorganisms and cells. In one embodiment, the devices comprise a container having an upper compartment and a lower compartment, wherein the upper compartment is connected to a passage through which a fluid sample can flow and enter the upper compartment, and wherein the lower compartment is connected to a passage through which a fluid sample can flow and exit the lower compartment; a mounting structure upon which a filter can be positioned; a filter, wherein the filter separates the upper compartment from the lower compartment, wherein the filter comprises a chromogen; a first template covering the filter, wherein the first template comprises a predetermined number of holes through which a liquid sample can flow through to discrete locations on the filter; and an absorbent pad under the filter and in contact with the filter.

Rapid bacterial quantification

The present invention provides devices and methods for determining the presence or absence of pathogens in a biological sample and for the concurrent determination of the antimicrobial susceptibility of pathogens present. The devices and methods of the present invention are able to make the determinations without pre-selection of target bacteria. Instead, all organisms from the sample are applied to the device. In wells where one or more antimicrobial agents are present only pathogens resistant to the antimicrobial agent grow. This allows the user to determine the presence of target pathogens and concurrently determine the resistance patterns of the organisms without the need to subculture the organisms. The present invention also provides methods of performing a business service of performing the determinations described above, and kits containing the devices and descriptions of the methods.

Methods and devices for the detection of pathogenic microorganisms and their antimicrobial susceptibility

The present invention relates to a method of detecting the presence of target microorganisms in a biological sample and of simultaneously determining the susceptibility of the microorganisms to antimicrobial agents. The target microbial organisms may be urinary pathogens. The methods include the steps of providing a multicompartment assay device with at least one compartment containing a medium capable of sustaining the growth of total viable microorganisms, at least one compartment containing a medium capable of sustaining the growth of target microorganisms, and at least one compartment containing an antimicrobial susceptibility interpretation medium. A biological sample is placed in each compartment and the presence and antimicrobial susceptibility of the target microorganisms which may be present is determined by analyzing which compartments exhibit microbial growth.

Optionally, an aminopeptidase inhibitor is added to the composition, to reduce endogenous enzymatic activity in samples. A method for use of the composition to detect the presence or amount of yeasts and/or molds in a test sample comprises inoculating a medium comprising the composition of claim 1 with said test sample; incubating the inoculated medium under conditions suitable for the growth of yeasts or molds; and, observing any detectable signal, whereby the presence of a detectable signal indicates the presence of yeasts or molds in the test sample.

A composition and method for detecting the presence or amount of yeasts and molds in a test sample is presented. The composition contains a substrate and an inhibitor for an aminopeptidase. The substrate has a signal moiety capable of providing a detectable signal when cleaved by an aminopeptidase in yeasts or molds. The aminopeptidase inhibitor serves to reduce the endogenous aminopeptidase activity in the test sample. The method to detect yeasts or molds in a sample includes inoculating a test sample with the disclosed composition, incubating the sample and observing any detectable signal that indicates the presence of yeasts or molds.
