The present invention concerns novel DNA compounds which function as transcriptional activating sequences. Recombinant DNA expression vectors which contain the transcriptional activating sequences and host cells transformed with these expression vectors are also provided. When properly positioned in an expression vector, the novel transcriptional activating sequences enable regulatable expression of an operably linked gene and enhance the structural stability of the expression vector.

The present invention comprises novel expression control sequences that promote transcription of DNA. The novel sequences have been positioned for expression of structural genes encoding bovine growth hormone derivatives on recombinant DNA expression vectors. Recombinant DNA expression vectors for use in Bacillus and other host cells are disclosed. The vectors comprise the veg promoter sequence of Bacillus subtilis, a novel ribosome binding site-containing sequence and a sequence that codes for a functional polypeptide. The present invention comprises improved derivatives of the tryptophan operon useful for expressing fused gene products in E. coli and relate organism. Two of the improved derivatives disclosed are encoded on 0.43 and 0.55 kb EcoRI restriction fragments from plasmids pCZ20 and pLEBGH2 (in strains NRRL B-15881 and NRRL B-15882), respectively. The modified derivatives have been placed on recombinant DNA cloning and expression vectors. A variety of expression vectors have been constructed that drive expression of fused gene products. Two novel gene sequences, encoding insulin-like growth factors I and II, have been expressed with the modified tryptophan operon system.
