Provided herein are compositions, kits, methods and devices for cyanide detection, particularly for cyanide detection in biological samples such as whole blood. The method comprises (1) contacting a sample with a cobinamide conjugate comprising a cobinamide moiety and a carrier; and (2) measuring the absorbance of light by the cobinamide conjugate. The present disclosure provides field-deployable cyanide detection methods, compositions, kits and devices, which provide rapid, accurate readout at the point of contact. Further provided herein is a method for determining exposure of a subject to cyanide.

Kits and methods for cyanide detection

A test device and method for determining the presence or absence of an analyte in a fluid sample, the test device including a support bearing a mark thereon, and a matrix defining an axial flow path. In operation, an observation area in the test device becomes transparent, thereby allowing the user to view a mark that is present on a support that is disposed beneath the observation area. Typically, the mark on the underlying support is configured as a minus (&#x2212;) sign. In the absence of analyte in the sample, the test device presents a negative result as a minus (&#x2212;) signal. In the presence of analyte in the sample, however, the mark operates in concert with a perpendicular test line on the observation area to present a positive result as a plus (+) signal that is visible to the user.

Lateral flow assay device

This invention provides solid phase specific binding lateral flow assay methods, devices and kits for quantitating high and low molecular weight analytes. The methods and devices of the invention employ labelled reagents which are either analyte analogs or complementary specific binding pair members for the analyte and a novel arrangement of capture zones comprising immobilized specific binding substances for either the analyte or the labelled reagent to effect bound from unbound labelled reagent as a function of analyte concentration. The capture zones are disposed on a non-bibulous matrix defining a flow path from a sample receiving zone to the capture zone. The devices of this invention also include multilane flow paths and multiple capture zones to quantitate analyte.

Quantitative lateral flow assays and devices

The efficiency and accuracy of one-step lateral flow assays can be improved by employing more efficient binding between participants in labeling and capture. Thus, in addition to analyte/anti-analyte interactions, specific binding is achieved through members of an irrelevant specific binding pair. Also included within the invention is a format wherein unlabeled competitor for analyte serves as a gatekeeper in the capture zone, competing with analyte for labeled anti-analyte, which analyte will be captured in a detecting portion of a capture zone.

One-step lateral flow assays

An improved one-step immunochromatographic assay which involves the binding of a predetermined amount of analyte to an antibody, enabling the control of the assay sensitivity, is disclosed. The system is especially useful as a controlled sensitivity fecal occult blood assay. Antibodies to a desired analyte, present at a predetermined concentration, are deposited on the sample pad of the reaction unit. The antibody binds analyte present in the sample, up to a threshold amount. Analyte which is present in the sample at a level above the threshold amount proceeds unbound through the sample pad and onto a membrane, where it reacts with an antibody-coated latex and a second, immobilized antibody to generate a positive signal.
