A kit for forming a biocompatible material provides a protein solution and a polymer solution including a derivative of a hydrophilic polymer with a functionality of at least three. Upon mixing. the protein solution and the polymer solution cross-link to form a non-liquid, three-dimensional network that degrades over time back to a liquid form. The polymer includes a degradation control region selected to achieve a desired degradation period and a cross-linking group selected to achieve a desired cross-linking period. The kit provides instructions for forming a mixture of the protein solution and polymer solution and for applying the mixture. The mixture serves as the foundation for multiple material composition species, each adapted to a specific therapeutic indication.

A delivery device applies a biocompatible and biodegradable barrier material to a tissue region, e.g., to seal a vascular puncture site. The material comprises two liquid components, which are pre-packaged in individual dispensers. Upon mixing, the liquid components cross-link to create a barrier matrix. A holder attaches to the delivery device. The holder mutually supports the first and second dispensers while the protein solution and polymer solution are conveyed from the dispensers into a fluid delivery channel. The protein and polymer solutions mix as a result of flow through the channel.

Systems for applying cross-linked mechanical barriers

A method is provided for the rapid formation of a biocompatible gel, and may be carried out in situ, i.e., at a selected site within a patient's body. The method involves admixing a biocompatible crosslinking component A having m sulfhydryl groups wherein m&gE;2 and a biocompatible crosslinking component B having n sulfhydryl-reactive groups wherein n&gE;2 and m&plus;n>4, wherein the sulfhydryl-reactive groups are capable of covalent reaction with the sulfhydryl groups upon admixture of the components under effective crosslinking conditions to form a gel in less than one minute. Suitable reaction conditions for carrying out the crosslinking reaction will depend on the particular components and the type of reaction involved; that is, the &ldquo;effective crosslinking conditions&rdquo; may involve reaction in bulk or in a solvent, addition of a base, and/or irradiation of the admixture in the presence of a free radical initiator. Exemplary uses include tissue augmentation, biologically active agent delivery, bioadhesion, and prevention of adhesions following surgery or injury. Reactive gel-forming compositions and systems are also provided.

Rapid-gelling biocompatible polymer composition and associated methods of preparation and use

A fluidic media introduction apparatus for closing an opening in a vessel wall includes an elongated member with a fluidic media introduction port and a guide wire lumen. The guide wire lumen extends from a proximal portion to a distal portion of the elongated member. A fluidic media mixing chamber is positioned in the elongated member and coupled to a fluidic media introduction port. A fluidic media delivery lumen is coupled to the mixing chamber. A fluidic media advancement port is positioned at the distal portion of the elongated member. An inflatable member is coupled to the distal portion of the elongated member.

Compositions, systems, and methods for creating in situ, chemically cross-linked, mechanical barriers

This invention relates generally to two-part polymer compositions that rapidly form covalent linkages when mixed together. Such compositions are particularly well suited for use in a variety of tissue related applications when rapid adhesion to the tissue and gel formation is desired. In particular, they are useful as tissue sealants, in promoting hemostasis, for drug delivery, in effecting tissue adhesion, in providing tissue augmentation, and in the prevention of surgical adhesions.
