The present invention provides a method for generating and isolating cell lines that functionally express molecular targets for drug discovery without utilizing information from the nucleic acid or amino acid sequence of the target protein. This procedure for the first time allows one to develop fast, high throughput screens for evaluation of test compounds that may modulate molecular targets whose specific nucleic acid or amino acid sequences are unavailable.

Method of screening for compounds that modulate the activity of a molecular target

Nucleic acid sequences, particularly DNA sequences, coding for all or part of a squalene synthetase, expression vectors containing the DNA sequences, host cells containing the expression vectors, and methods utilizing these materials. The invention also concerns polypeptide molecules comprising all or part of a squalene synthetase, and methods for producing these polypeptide molecules.

Two related, novel antibiotic substances, peptifluorin and neopeptifluorin, are prepared by cultivation of strains of Pseudomonas fluorescens, A. T. C. C. No. 53,958 and A. T. C. C. No. 55,129, respectively.

Cultivation of a strain of the microorganism, Preussia sp., that has been deposited in the American Type Culture Collection as A.T.C.C. No. 20,923, yields a novel antibiotic substance, culpin, that has activity against a selected spectrum of microorganisms.
