A method of manipulating and/or investigating cellular bodies () is provided. The method comprises the steps of: providing a sample holder () comprising a holding space () for holding a fluid medium (); providing a sample () comprising one or more cellular bodies () in a fluid medium () in the holding space (); generating an acoustic wave in the holding space exerting a force (F) on the sample () in the holding space (). The method further comprises providing the holding space () with a functionalised wall surface portion () to be contacted by the sample () and the sample () is in contact with the functionalised wall surface portion () during at least part of the step of application of the acoustic wave. A system and a sample holder () are also provided.

Method and system for studying biological cells

A method of excitation microscopy, in particular STimulated Emission Depletion (STED) microscopy, ins provided which comprises: providing a sample; trapping an object in the sample at a trapping position, in particular by applying a position dependent trapping force to the object; positioning, in particular focusing, a depletion beam at an interaction position in the sample for illumination of a portion of the sample associated with the trapped object. The method comprises at least one of controlling the depletion beam such that, at least when the depletion beam is positioned at the interaction position, an optical force exerted by the depletion beam on the object causes a displacement of the object less than the optical resolution, preferably less than half the optical resolution of an imaging system for observing a STED fluorescence; and controlling at least one of the depletion beam and the trapping force on the object such that, at least when the depletion beam is positioned at the interaction position, an optical force exerted by the depletion beam on the object is less than 5% of the trapping force, preferably less than 3%, more preferably less than 1%. An according system is also provided.

Microscopy method and system

A molecular manipulation system for investigating molecules, having a sample holder constructed to hold a sample comprising a plurality of molecules attached on one side to a surface in the sample holder and on another side attached to a microbead of a plurality of microbeads. The system having; an acoustic wave generator to generate an acoustic wave exerting a force on the microbeads in the sample; and a detector device to detect a response of the plurality of microbeads in the sample on the force exerted by the acoustic wave to investigate the molecules attached to the microbeads.

Molecular manipulation system and method

The present disclosure concerns a method and system for imaging a molecular strand (MS). The method comprises providing a sample volume (SV) comprising the strand (MS); providing an excitation beam (EB) having an excitation focus (EF) in the sample volume (SV); scanning the excitation focus (EF) in the sample volume (SV) along a one dimensional scanning line (SL); trapping an end of the strand (MS) in the sample volume (SV) and extending the strand (MS) along a one-dimensional trapping line (LL) parallel to the scanning line (SL); aligning the trapping line (LL) to coincide with the scanning line (SL) to have the scanning excitation focus (EF) coincide with the strand (MS); and recording the fluorescence response (FR) as a function of a plurality of distinct scanning positions (X) of the excitation focus (EF) along the scanning line (SL).
