An integrated bioreactor and reseeding chamber system and a separate bioreactor and complimentary reseeding chamber system comprising a bioreactor and a separate reseeding chamber. The bioreactor comprises a bioreactor chamber; a first groove attached to the bioreactor chamber; a second groove removably attached to the bioreactor chamber. A first bar and a second bar may be removably inserted into the first groove and the second groove respectively. Each of the first bar and the second bar may have at least an oval hole. At least a crossbar is attached substantially perpendicularly to the first bar and the second bar to form a crossbar-bars construct through the oval holes. A knob may be installed on each of the crossbars. The reseeding chamber may comprise dividers fixedly attached to a bottom of the reseeding chamber.

Bioreactor and reseeding chamber system and related methods thereof

The present invention provides a bioreactor controller with a simple user interface comprising a microcontroller and a linear motor. The bioreactor controller is microcontroller-based, has a greater temporal accuracy, and inexpensive. The microcontroller of the bioreactor controller selects a protocol mode and a setup mode by the user, runs the protocol mode if the setup mode is not selected by the user, stops running the protocol mode for a predetermined time if the protocol mode is selected by the user until the user is completed with calculations and record-keeping, detects movement error of the bioreactor controller, and informs the error to the user.

Bioreactor controller device and related method thereof

Provided herein are methods of culturing organized skeletal muscle tissue from precursor muscle cells by cyclically stretching and relaxing said muscle cells on a support in vitro for a time sufficient to produce said organized skeletal muscle tissue, including reseeding said organized skeletal muscle tissue by contacting additional precursor muscle cells to said organized skeletal muscle tissue on said solid support, and then repeating said step of cyclically stretching and relaxing said muscle cells in said support in vitro for time sufficient to enhance the density (i.e., increased number of nuclei and/or number of multinucleated cells) of said organized skeletal muscle tissue on said support.

Methods for making a tissue engineered muscle repair (TEMR) construct in vitro for implantation in vivo

Provided herein are scaffolds and methods useful to promote the formation of functional clusters on a tissue, for example, motor endplates (MEPs) or a component thereof on skeletal muscle cells or tissue, as well as the use of scaffolds so produced for repairing a tissue injury or defect.

Bioscaffolds for formation of motor endplates and other specialized tissue structures

The disclosure provides improved materials and methods for optically clearing biological tissue that is subsequently used for deep tissue imaging analysis. Also provided is a description of a microscopic image acquisition methodology in which imagery of intact tissues are acquired to rapidly acquire microscopy data on a whole-organ scale to maximize cost effectiveness for biological microscopy and minimize time spent performing such analysis.

Compositions and methods for clearing a biological sample

A method of producing organized skeletal muscle tissue from precursor muscle cells in vitro comprises: (a) providing precursor muscle cells on a support in a tissue media; then (b) cyclically stretching and relaxing the support at least twice along a first axis during a first time period; and then (c) optionally but preferably maintaining the support in a substantially static position during a second time period; and then (d) repeating steps (b) and (c) for a number of times sufficient to enhance the functionality of the tissue formed on the support and/or produce organized skeletal muscle tissue on the solid support from the precursor muscle cells.

Methods are provided for producing a bioscaffold from natural tissues by oxidizing a decellularized tissue to produce a bioscaffold having pores therein. The pore size and porosity is increased to better accommodate intact cells so that live cells can better infiltrate and inhabit the bioscaffold. The bioscaffold may be freeze-dried or lyophilized, sterilized and (optionally) aseptically packaged for subsequent use. A further aspect of the present invention is a bioscaffold produced by the processes described herein. Methods of treatment using the bioscaffold as a graft or as a biomedical implant for implantation are also provided. Also provided are methods of seeding a bioscaffold with mammalian cells, wherein the seeding carried out either in vitro or in vivo, and wherein a bioscaffold produced as described herein is utilized for said seeding.
