Presented herein are compositions, systems, and methods related to optical substrates that simultaneous (1) enhance a fluorescence signal emitted by a fluorophore and (2) enhance 'contrast' signal that comprises scattered signal intensity over substrate reflectivity at a non-fluorescent wavelength. In certain embodiments, the optical substrate comprises a thin, transparent, dielectric layer. In alternative embodiments, the optical substrate comprises a stack of thin, transparent dielectric layers, for example, that is designed for both specific scattering enhancement and fluorescence enhancement.

Compositions, systems, and methods for enhanced label-free and fluorescence-based detection of nanoparticles

A spectral reflectance imaging device for detecting nanoparticle exosome biomarker targets includes an illumination source that illuminates a substrate with a plurality of separate wavelengths of incoherent light. The substrate includes an oxide layer and a binding agent to selectively bind nanoparticle exosome biomarker targets to the substrate. An imaging device bindings the light reflected from or transmitted through the substrate and an image processing system detects the nanoparticle exosome biomarker targets a function of the change in reflective properties of the substrate.

Detection of exosomes having surface markers

Provided herein are methods for capturing extracellular vesicles from a biological sample for quantification and/or characterization (e.g., size and/or shape discrimination) using an SP-IRIS system. Also provided herein are methods of detecting a biomarker on captured extracellular vesicles or inside the captured vesicles (e.g., intra-vesicular or intra-exosomal biomarkers).

Multiplexed phenotyping of nanovesicles

An imaging system uses polarized light to illuminate the target and then uses a polarization filter to remove the light that is reflected from the target without modification. The target can include one or more anisotropic objects that scatter the light and alter the polarization state of the reflected light and causing it to be selectively transmitted to the imaging device which can record the transmitted light through the filter. The illuminating light can be circularly polarized and the filter can remove the circularly polarized light. The target can include asymmetric nanoparticles, such as nanorods that alter the amplitude or phase of the scattered light enabling pass through the filter to be detected by the imaging device.

Polarization enhanced interferometric imaging

A system for analyzing one or more liquid samples includes a microwell plate including a plurality of rows of wells configured to store liquid samples, a sensor array that is moveable relative to the microwell plate along a first axis between a first position and a second position to allow a portion of the sensor array to be disposed within a first one of the plurality of rows of wells when the sensor array is in the second position, an objective, and one or more linear translation stages configured to move the microwell plate relative to the objective (i) along a second axis that is orthogonal to the first axis, (ii) along a third axis that is orthogonal to the first axis and the second axis, or (iii) both (i) and (ii).

Systems and methods for imaging microwell plate samples

Systems and methods for high-throughput processing of assay plates include a calibration nanoparticle to facilitate automated focusing of the imaging system. An assay plate includes a base layer, a transparent layer in contact with the base layer, and at least one calibration nanoparticle having a pre-defined size immobilized on the assay plate surface. The assay plate surface can be functionalized to selectively bind to biological targets. The assay plate can be used in an imaging system for high-throughput autofocus and biological target detection.
