Disclosed are 6.alpha.-derivatized estriol compounds which, when conjugated to a protein, are useful in the in vivo preparation of antibodies specific to estriol. When labeled with a detectable label, the estriol derivatives are useful as haptens in a competitive immunoassay for estriol which demonstrate superior sensitivity with respect to estriol specific antibodies.

Synthesis of 6.alpha.-functionalized estriol haptens and protein

The present invention involves the synthetic conversion of pyridinoline (PYD) to deoxypyridinoline (DPD) via an intermediate ester which is subsequently cleaved. The transformation of PYD into a xanthate-type ester intermediate facilitates the subsequent removal of the substituted hydroxyl functionality thus providing the desired DPD. The method provides a new chemical pathway for the preparation of DPD, resulting in significantly increased yields as compared to traditional methods of product isolation.

Conversion of pyridinoline to deoxypyridinoline

The determination of human serum albumin (HSA) in urine is carried out by contacting the urine with a reagent system containing a buffer and a protein error indicator. Other proteins normally found in urine can compete with the HSA for interaction with the protein error indicator thereby affecting the sensitivity of the test for HSA. The present invention involves the addition of certain polymeric competitive inhibitors to the reagent system which inhibit the interaction of the urinary proteins other than HSA with the protein error indicator to a greater extent than the interaction with HSA is inhibited thereby increasing the reagent's sensitivity for HSA. Suitable polymeric inhibitors include poly(vinyl alkyl esters), poly(alkyl acrylates), poly(vinyl alkyl carbonates) and poly(vinyl alkyl ketones).

Method for the detection for protein

Disclosed are novel folic acid-alkaline phosphatase conjugates useful in a competitive immunoassay for folic acid in a biological fluid such as serum. A preferred conjugate involves a folate hapten bound to (N-Succinimidyl�4-iodoacetyl!aminobenzoate) through a 1,12-diamino-4,9-dioxadodecanyl spacer arm.

A test device for determining the presence of leukocyte cells, esterase or protease in a test sample is disclosed. The test device comprises a carrier matrix having a reagent composition incorporated therein, wherein the reagent composition comprises a lactate ester having the structure ##STR1## wherein A is an alcohol blocking group, and wherein B--O-- is a residue of compound B--OH and provides a detectable response when the lactate ester is hydrolyzed; and a buffer. In a preferred embodiment, the lactate ester has the structure ##STR2## wherein X is O, S or NR.sup.2, R is aryl or lower alkyl, R.sup.1 is hydrogen or lower alkyl, and R.sup.2 is hydrogen, lower alkyl or aryl.

A composition and method for determining the presence of leukocyte cells, esterase or protease in a test sample are disclosed. The composition contains a lactate ester having the structure ##STR1## wherein A is an alcohol blocking group, and wherein B-O- is a residue of compound B-OH and provides a detectable response when the lactate ester is hydrolyzed; and a buffer. One class of lactate ester has the structure ##STR2## wherein X is O, S or NR.sup.2, R is aryl or lower alkyl, R.sup.1 is hydrogen or lower alkyl, and R.sup.2 is hydrogen, lower alkyl or aryl.
