Systems and methods for particle sorting are presented including a monitoring system downstream of a particle separator or sorter. The system can utilize the monitoring system to adjust or calibrate operational parameters of the system in real time. When a particle of interest is mis-sorted, the probability is high that the particle of interest has been sorted into a non-targeted sortable unit that was adjacent in sequence to the sortable unit that was expected to include the particle of interest. The monitoring system monitors non-targeted sortable units in the system that were adjacent in sequence to targeted sortable units that are predicted to contain particles of interest. Signals from the monitoring system enable automated adjustment or calibration of operational parameters of the system such as sort delay or purity mask parameters.

Apparatus and methods for detecting, characterizing, and compensating motion-related error of moving micro-entities are described. Motion-related error may occur in streams of moving micro-entities, and may represent a deviation in and expected arrival time or an uncertainty in position of a micro-entity within the stream. Motion-related error of micro-entities is observed in a flow cytometer, e.g., as pulse jitter, and is found to have a functional dependence on a parameter of the system. The pulse jitter can be compensated, according to one embodiment, by adjusting data acquisition observation windows. For the flow cytometer, a reduction of pulse jitter can improve measurement accuracy, resolution of doublets, system throughput, and enable an increase in an interrogation region for probing the micro-entities.

A high speed fluid switch is used to separate desired particles from a source stream containing particles, such as cells. A droplet generator creates and regulates a stream of droplets, which intersects with a source stream containing the particles to be sorted. The stream of droplets can be regulated such that portions of the source stream containing desired particles are deflected into a collection vessel, or such that portions of the source stream containing desired particles are permitted to pass uninterrupted. The high speed fluid switch may be implemented on a microfluidic chip.

The various embodiments disclosed herein utilize multiple lasers that have different wavelengths and a single detection path. The lasers are mounted orthogonal to one another so that one laser will provide a forward angle light scatter (FALS) signal in the detection path, and one laser will provide a side scatter signal in the detection path (i.e., the single detection optics are approximately in-line with the FALS laser and approximately orthogonal to the side scatter laser). The single detector path spectrally separates the forward and side scatter signals prior to applying them to their respective detectors for measurement.
