The present invention is directed to a phospholipid-based NIR molecular beacon, having a phospholipid moiety; with an NIR fluorophore moiety covalently linked to a phospholipid glycerol backbone and a quencher moiety covalently linked to the phospholipid glycerol backbone. Additionally, provided herein is methods of analyzing a sample for the presence of a phospholipase and methods of identifying the activity of a phospholipase in vivo utilizing phospholipid-based NIR molecular beacon.

Vivo detection of phospholipase activation

The application relates to a nanovesicle comprising a bilayer of at least 15 mol % porphyrin-phospholipid conjugate, wherein the conjugate comprises a porphyrin, porphyrin derivative or porphyrin analog covalently attached to the phospholipid side chain. The nanovesicle can be used for photothermal therapy, photoacoustic imaging, and fluorescence imaging. The application also discloses a method of preparing the said nanovesicle.

Porphyrin nanovesicles

There is herein described a nanovesicle comprising a bilayer of porphyrin-phospholipid conjugates. Each porphyrin-phospholipid conjugate comprises one porphyrin, porphyrin derivative or porphyrin analog covalently attached to a lipid side chain at one of the sn-1 or the sn-2 positions of one phospholipid. Further, the nanovesicle has a defined regioisomeric ratio of sn-1:sn-2 porphyrin-phospholipid conjugates.

Method for the synthesis of porphyrin-phospholipid conjugates

The present invention relates to the field of conjugates comprising a substrate which is attached to at least one photoactivatable killing agent and at least one first quencher, and methods for their use. More particularly, the present invention relates to photodynamic therapy agents. The invention further relates to methods for decontaminating blood and methods for treating cancer or viral infection in a subject using the conjugates of the present invention.

Activatable photodynamic therapy agents

The present invention relates to novel antineoplastic agents and cancer diagnostic agents that specifically target neo-plastic cells via the GLUT transportation system. More specifically, the invention relates to conjugates of 2-deoxyglucose, wherein a linker, which includes a covalent bond, is attached to 2-deoxyglucose at the 2 position, and the linker is attached to a therapeutic or diagnostic agent. The invention also relates to methods of treating tumor disease and methods of making the novel compounds of the present invention. The agents of the present invention are superior to previous agents as they are targeted via GLUT transporters.
