Described herein are methods of treating, diagnosing, and/or prognosing a disease in a subject relating to detection of the glycosylation state of the antibodies present in the subject. In some embodiments, the disease can be an infection. In some embodiments, an antibody glycosylation state that is indicative of the presence of a disease, or a need for treatment of a disease can be reduced glycosylation (e.g., galactosylation, sialation, fucosylation, and/or afucosylated branched glycoforms).

Methods of diagnosis and treatment of tuberculosis and infection

Systems and methods have been developed to design and engineer glycan dynamics to improve immunogen antigenicity. These include systems for identify glycosylation sites that that impact binding of antibodies to the immunogen, and modifying the glycan profiles on these glycosylation sites to synthesize novel immunogens, antibodies and vaccines. Then, the machine learning algorithm may output data relating to the glycosylation sites that are determinant or likely impact the binding affinity of the variants to the one or more antibodies.

Synthesizing vaccines, immunogens, and antibodies

In accordance with an embodiment of the invention, there is provided a method for: a) high-throughput, multiplexed, affinity-based separation of proteins&#x2014;especially low abundance proteins&#x2014;from complex biological mixtures such as serum; and b) high-throughput, multiplexed, affinity-based separation of cells&#x2014;especially rare cells&#x2014;from complex biological mixtures such as blood or blood fractions. The separation of proteins or cells is achieved based on differential binding to affinity-capture beads of different sizes and then sorting the protein-bound or cell-bound beads using the concept of centrifugal-induced Dean migration in a spiral microfluidic device. This method enables continuous-flow, high throughput affinity-separation of milligram-scale protein samples or millions of cells in minutes after binding. This is particularly applicable to the isolation of antigen-specific antibodies from polyclonal sera and antigen-specific immune cells or circulating tumor cells from blood, both of which are otherwise highly labor-intensive and expensive to perform.
