Polypeptides belonging to the family of late proteins of the cornified envelope (LCE), fragments of the polypeptide, isolated nucleotide sequences encoding the polypeptides, and cosmetic and/or pharmaceutical compositions containing such polypeptides are described. The polypeptides have cosmetic and/or therapeutic use to reinforce the barrier function of the epidermis, prevent and/or treat the signs of skin dryness and prevent and/or treat disorders of the barrier function or the weakening of the epidermis.

Polypeptide expressed in the stratum corneum and use thereof

An in vitro method for screening for candidate compounds for preventing and/or attenuating skin ageing, and/or hydrating skin, includes: a) contacting a test compound with a sample of papillary fibroblasts; b) measuring the expression of a gene selected from PDPN, CCRL1 and NTN1, in the papillary fibroblasts; and c) selecting compounds for which an activation of at least 1.5 fold of the expression of at least one of the genes is measured in the treated papillary fibroblasts compared with untreated papillary fibroblasts. Another in vitro method includes: a) contacting a test compound with a sample of reticular fibroblasts; b) measuring the expression of a gene selected from MGP, PPP1R14A and TGM2, in the reticular fibroblasts; and c) selecting compounds for which an activation of at most 1.0 fold of the expression of at least one of the genes is measured in the treated reticular fibroblasts compared with untreated reticular fibroblasts.

Markers of papillary and reticular fibroblasts and uses thereof

Identification and use of compounds which inhibit the expression or activity of micro-RNAs for preventing and/or attenuating ageing. An in vitro method for screening for candidate compounds for preventing and/or attenuating ageing of the skin including (a) bringing at least one test compound in contact with a sample of fibroblasts, (b) measuring the expression or the activity of at least one microRNA chosen from miR-134 and miR-152 in said fibroblasts, and (c) selecting the compounds for which an inhibition of at least 20%, preferably at least 30%, preferably at least 40% of the expression or an inhibition of at least 20%, preferably at least 30%, preferably at least 40% of the activity of at least one microRNA is measured in the fibroblasts treated in (a) compared with the untreated fibroblasts.

Inhibitors of micro-RNAs for use for preventing and/or attenuating skin ageing

At least one microRNA selected from among hsa-miR-330, hsa-miR-7 and hsa-miR-137, the mature forms thereof, and the precursors thereof, for depigmenting the skin, and an in vitro method for identifying depigmenting compounds, which includes the steps of: a) placing at least one test compound in contact with a sample of melanocytes; b) measuring the expression or activity of at least one microRNA selected from among miR-330, miR-7 and miR-137, the mature forms thereof, and the precursors thereof, in the melanocytes; c) selecting the compounds for which at least 20% activation of the expression or of the activity of at least one of the microRNAs is measured in the melanocytes treated in a) by an element for comparing with the untreated melanocytes.

The polypeptides have cosmetic and/or therapeutic use to reinforce the barrier function of the epidermis, prevent and/or treat the signs of skin dryness and prevent and/or treat disorders of the barrier function or the weakening of the epidermis.

The present invention relates to novel peptide compounds having the following formula [RRN&#x2014;CH(Ra)&#x2014;CO]-(AA)-(AA)-X&#x2014;X&#x2014;X&#x2014;Y&#x2014;Y&#x2014;Y&#x2014;X-(AA)-[NH&#x2014;CH(Rb)&#x2014;COOR].
