Microbial process for preparing pravastatin

The invention refers to a new microbial process for preparing .beta.,.delta.-dihydroxy-7-[1,2,6,7,8,8a-hexahydro-2.6-dimethyl-8-(2-methy l-butyryloxy)-naphthalen-1-yl]-heptanoic acid .delta.-lactone of formula (I) and .beta.,.delta.-dihydroxy-7-[1,2,6,7,8,8a-hexahydro-2.6-dimethyl-8-(2-methy lbutyryloxy)-naphtalen-1-yl]-heptanoic acid of formula (II), ##STR1## by the aerobic fermentation of the submerged culture of an imperfect fungus strain biosynthesizing the above compounds, in a nutrient medium containing utilizable carbon and nitrogen sources as well as mineral salts, and by isolating the product of formula (I), which comprises cultivating a strain of the new Aspergillus obscurus fungus species producing the above compound(s) of formulas (I) and/or (II), in a temperature range of 25.degree. to 30.degree. C. and, if desired, separating the product(s) formed from the fermentation broth, then isolating it in the lactone form of formula (I) and, if desired, purifying the same.

The invention relates to a process for the production of 9.alpha.-hydroxy-4-androstene-3,17-dione from natural sterols of plant or animal origin or from mixtures thereof by the submerged aerobic fermentation of an enzyme-deficient, sterol-de-grading microorganism in a nutrient medium containing utilizable carbon and nitrogen sources as well as mineral salts, and by isolating the product formed, which comprises transforming the sterol or sterol mixture with the culture of a 1,2-steroid-dehydrogenase deficient strain of new Mycobacterium roseum species which is able to degrade the side-chain of natural sterols, preferably with Mycobacterium roseum sp. nov. 1108/1 deposited at the National Collection of Agricultural and Industrial Microorganisms, Budapest, Hungary under the number NCAIM B (P) 000339, and by isolating and purifying the 9.alpha.-hydroxy-4-androstene-3,17-dione obtained.

These compounds can be prepared by removing the ester group and the p-nitrobenzyloxycarbonyl protective group of a 9.alpha.,11.alpha.,15-trihydroxy-16-p-nitrobenzyloxycarbonylamido-17-pheny l-5-cis,13-trans-18,19,20-trinor-prostadienoic acid derivative of general formula XII--wherein C-15 and C-16 may have either S or R configuration, W is as defined above, and Y stands for a lower alkyl group--in an optional sequence with the limitation that in those compounds of general formula I where W is as defined above and Y stands for a lower alkyl group, solely the p-nitrobenzyloxycarbonyl group is removed, and the resulting product of general formula I is optionally converted with an organic or inorganic acid into a salt. The new prostaglandin derivatives of the invention have valuable therapeutical properties, and can be applied in cattle raising for estrus and birth synchronization, furthermore in the veterinary praxis for the treatment of sterility, chronic endometritis and pyometry.

The invention relates to new 16-amino-prostaglandin derivatives of general formula I, ##STR1## wherein C-15 and C-16 may have either S or R configuration, Z stands for a hydrogen atom or a lower alkyl group, and their acid addition salts. These compounds can be prepared by removing the ester group and the p-nitrocarbobenzyloxy protective group of a 9.alpha., 11.alpha., 15-trihydroxy-16-p-nitrobenzyloxycarbonylamido-5-cis,13-trans-prostadienoi c acid derivative of general formula XII--wherein C-15 and C-16 may have either S or R configuration and Z stands for a lower alkyl group--in an optional sequence with the limitation that in those compounds of general formula I where Z stands for a lower alkyl group solely the p-nitrocarbobenzyloxy group is removed, and the resulting product of general formula I is optionally converted with an organic or inorganic acid into a salt. The invention provides a process for the preparation of antibiotic-producer Micromonospora strains having modified genetic material, wherein a protoplast suspension is prepared with lysozyme, under osmotically buffered conditions ensured by sucose, from each culture of the two genetically marked mutants of the antibiotic-producer Micromonospora strain following cultivation in a glycine medium, both suspensions obtained are combined in the presence of polyethylene glycol, and incubated at room temperature, the resulting fused protoplasts are suspended in soft agar, then plated on an agar plate containing sucrose, prolin and inorganic salts, incubated for 20 to 30 days, and finally all mutants are selected from the regenerated colonies which are sure to have genetic material different from that of the parent strains. The process can be advantageously applied to improve productivity of antibiotic-producer strains having industrial importance.
