The present invention is related to an holographic probe device for recording a gabor hologram comprising: a coherent optical fiber bundle comprising a distal end and a proximal end; a recording medium optically coupled to the proximal end of the coherent optical fiber bundle; a light source producing in use a single light beam, illuminating the distal end of the coherent optical fiber bundle and the object to be observed.

Method for recording gabor hologram

The present invention is related to a method for characterizing transparent objects () in a transparent medium (), said transparent objects () presenting an optical focal area () said method comprising the steps of: illuminating a sample comprising the objects () to be characterized by means of a directional light () source, thereby inducing light intensity peaks () at the focal area of said transparent objects; determining at least one characteristic of the light intensity peak () induced by said object to be characterized, determining from said light intensity peak () at least one property of said object.

Optical method for characterizing transparent particles

An Interferometer for off-axis digital holographic microscopy includes a recording plane, and a grating located in a plane optically conjugated with the recording plane. The grating defining a first and a second optical path, the optical path corresponding to different diffraction order.

Off-axis interferometer

A method for detecting a three dimensional object smaller than 300 nm includes providing a light source producing a first light beam (), the light source being at least partially coherent; and splitting the first light beam () into an object beam () and a reference beam () by a first beam splitter (BS). An image of the light source is produced in a light source image plane by a first microscope objective (L), in the optical path of the object beam (). The three dimensional object to be detected in an object cell () is positioned in the optical path of the object beam (), between the first beam splitter (BS) and the first microscope objective (L). The object beam () and the reference beam () are recombined into a recombined beam () by use of optical devices. An optical stop () is placed in the light source image plane of the microscope objective (L) on the optical axis of the microscope objective (L). Interferometric signals produced by the interaction between the reference beam and the object beam are recorded with a recording device (). The recombined beam () is focused onto the recording device () with a focuser (L). A three dimensional picture of the three dimensional object is reconstructed to be detected from the interferometric signal, thereby detecting the three dimensional object.

Holographic microscopy and method to investigate nano-sized objects

The present invention is related to a compact microscope able to work in digital holography for obtaining high quality 3D images of samples, including fluorescent samples and relatively thick samples such as biological samples, said microscope comprising illumination means () at least partially spatially coherent for illuminating a sample () to be studied and a differential interferometer () for generating interfering beams from said sample () on the sensor () of an electronic imaging device(), said interferometer () comprising namely tilting means () for tilting by a defined angle one the interfering beams (or) relatively to the other, said tilting resulting into a defined shift () of said interfering beam on the sensor of the electronic imaging device (), said shift () being smaller than spatial coherence width of each beam, said microscope being able to be quasi totally preadjusted independently from the samples so that minimum additional adjustments are required for obtaining reliable 3D images of samples.

Digital holographic microscope for 3D imaging and process using it

A method and device for obtaining a sample with three-dimensional microscopy, in particular a thick biological sample and the fluorescence field emitted by the sample. One embodiment includes obtaining interferometric signals of a specimen, obtaining fluorescence signals emanating from the specimen, recording these signals, and processing these signals so as to reconstruct three-dimensional images of the specimen and of the field of fluorescence emitted by the specimen at a given time. Another embodiment includes a digital holography microscope, a fluorescence excitation source illuminating a specimen, where the microscope and the fluorescence excitation source cooperate to obtain interferometric signals of the specimen and obtain fluorescence signals emanating from the specimen, means for recording the interferometric signals and fluorescence signals, and means for processing the interferometric signals and the fluorescence signals so as to reconstruct three-dimensional images of the specimen and of the field of fluorescence emitted by the specimen at a given time.

Method and device for obtaining a sample with three-dimensional microscopy

A method measures the three dimensional position of particles in a fluid medium contained in a sample by recording a digital hologram of the particles on an image sensor and by reconstructing the image of the particles from the hologram. The recording includes providing a source beam with a coherent source, and generating at least two beams from the source beam, namely a reference beam and at least one object beam, wherein the reference beam and the object beam are mutually coherent. The sample is illuminated by condensing the object beam onto the sample in order to obtain a scattered object beam for each particle. The scattered object beam is scattered into a spherical converging object beam toward the image sensor for each particle. Further, the recording includes forming a diverging spherical beam from the reference beam, and superposing the spherical converging object beam for each particle and the diverging spherical beam on the image sensor in order to obtain on the image sensor an hologram of the particles by interfering the beams.
