A pentamethine cyanine fluorescent dye having the general formula I, a synthetic method and a use thereof are disclosed. In the general formula, X is CHO or CHCRR; Rand Rare independently selected from the group consisting of (CH)Rand et al.; Rand Rare independently selected from the group consisting of CN, COOH and COOR; R, Rand Rare independently selected from the group consisting of H, SORand COOR; Ris H or Calkyl; Ris H or CH; Ris N(RRRR); Ris Calkyl; R, R, R, Rand Rare independently selected from the group consisting of H, Calkyl, (CH)ORand (CHRCHO)R; Yis halogen anion or OTs; and n, m and p are integers of 0-18. The dye can be utilized to sensitively detect viscosity of micro-environment such as tissues and cells.

The present invention relates to compounds shown in the general formula (I) or (III), the salts thereof or their mixtures, as well as their preparation method and application. In the general formula (I), Xis H or COH; Xis OH or phenyl group with 0-2 sulfonic acid substituents, and the sulfonic acid substituents are located at random positions of a benzene ring; when Xis OH, Xis H; when Xis phenyl group with 0-2 sulfonic acid substituents, Xis H or COH; n is an integer of 0-2; and in the general formula (III), n and m are respectively an integer of 0-2. The compounds and the mixtures not only have improved light resistance, ozone resistance and water resistance, but also have excellent water solubility and long-term stability in ink-jet ink.

The dye reacts with biosubstrate including protein, liposome and DNA, and shows excellent selectivity. It has good living cell membrane permeability and shows good selectivity in living cell staining In addition, the dye has good stability and low background fluorescence, and can be used together with other fluorescent dyes having consistent wavelengths for single-channel excitation and multi-channel detection. The dye can be used in the fields of protein labeling and detecting, fluorescence immunoassay and living cell selective imaging and the like.
