Disclosed are a novel antibody specifically binding to the tumor-immunosuppressant, TIGIT (T cell immunoglobulin and immunoreceptor tyrosine-based inhibitory motif [ITIM] domain) or an antigen-binding fragment thereof, a nucleic acid encoding the antibody or the antigen-binding fragment thereof, a vector and a host cell including the nucleic acid, a method for producing the antibody or the antigen-binding fragment thereof, a pharmaceutical composition containing the antibody or the antigen-binding fragment thereof as an active ingredient, and uses of the pharmaceutical composition. The antibody specifically binding to TIGIT or the antigen-binding fragment thereof and the pharmaceutical composition containing the same as an active ingredient are preferably used for the treatment of cancer or tumors.

Anti-TIGIT antibodies and uses thereof

The present invention relates to a mesenchymal stem cell having immunomodulatory activity and a preparation method therefor and, more specifically, to: a rapamycin-treated mesenchymal stem cell having immunomodulatory activity, which expresses any one or more cell surface factors selected from the group consisting of CCR1, CCR2, CCR3, CCR4, CCR7, CCR9 and CXCR4; a cell therapy composition comprising the mesenchymal stem cell, for preventing or treating immune disorders; and a preparation method for the mesenchymal stem cell having immunomodulatory activity. The rapamycin-treated mesenchymal stem cell having immunomodulatory activity, according to the present invention, has increased expression of IDO, TGF-&#x3b2; and IL-10 which are factors having immunomodulatory activity, has decreased expression of Phospho-mTOR, Rictor and Ractor which are signal transduction factors of mTOR, and has increased expression, in the cell, of autophagic inducer Beclin1, ATG5, ATG7, LC3I or LCII. If this cell is used as a cell therapy in individuals having immune disorders, it is possible to effectively treat immune disorders.

The daurinol compound according to the present invention exhibits excellent therapeutic effects on arthritis and inflammatory bowel diseases by having an excellent activity in inhibiting the generation of IL-17 and TNF-&#x3b1;, which are inflammatory cytokines causing inflammation, and can alleviate transplant rejection by inhibiting the proliferation of alloreactive T cells and increasing regulatory T cells; therefore, the daurinol compound according to the present invention can be used as a pharmaceutical composition capable of treating and preventing immune diseases. In addition, being a natural substance derived from the Mongolian medicinal plantor, the daurinol compound according to the present invention does not cause cytotoxicity and thus may be used as a composition for a functional health food capable of preventing and remedying immune diseases.

Composition for treating immune diseases containing daurinol compound as active ingredient

A method for manufacturing a porous ceramic scaffold having an organic/inorganic hybrid coating layer containing a bioactive factor includes (a) forming a porous ceramic scaffold; (b) mixing a silica xerogel and a physiologically active organic substance in a volumetric ratio ranging from 30:70 to 90:10 and treating by a sol gel method to prepare an organic/inorganic hybrid composite solution; (c) adding a bioactive factor to the organic/inorganic hybrid composite solution and agitating until gelation occurs; and (d) coating the porous ceramic scaffold with the organic/inorganic composite containing the bioactive factor added thereto. In accordance with the method, the porous ceramic scaffold may be uniformly coated with the organic/inorganic hybrid composite while maintaining an open pore structure, and stably discharge the bioactive factor over a long period of time.
