Provided are a method of screening an inhibitor of caspase activity by lipopolysaccharide and a method of screening a therapeutic agent for inflammatory diseases or sepsis using the same. Accordingly, it is possible to develop a caspase-4-specific inhibitor.

The present invention relates to a method for analyzing the interaction between HIF-1 peptide and VBC protein using fluorescence polarization, more precisely, a method for quantitative analysis of formation of HIF-1-VBC protein complex which is composed of the steps of 1) preparing a fluorescent probe by attaching a fluorescein to hydroxyproline containing HIF-1 peptide; 2) reacting the fluorescent probe with VBC protein; and 3) measuring the fluorescence polarization of the above reactant and then comparing the fluorescence polarization with that of the fluorescent probe itself to investigate the changes of fluorescence polarization; a method for screening an inhibitor of the binding of HIF-1 peptide and VBC protein using the above method; and a method for analyzing the activity of prolyl hydroxylase using the above method. The method of the present invention enables simple analysis of the interaction between HIF-1 peptide and VBC protein by observing changes of fluorescence polarization and thus, it can be effectively used for high-speed screening using a well plate.

A method and a kit for detecting a target protein in a sample with a signal amplification strategy are provided. The signal amplification strategy is established for the aptamer-based molecular recognition of a target protein with concomitant release of single-stranded DNA (G-DNA), which binds complementarily to a single-stranded RNA comprising a fluorophore and a quencher ('F-RNA-Q'). The fluorescence-quenched RNA is then degraded by RNase H to result in a fluorescence signal, and the undamaged G-DNA is recycled to yield fluorescence amplification.

Disclosed relates to a method and an apparatus for measuring fluorescence polarization FP in a lab-on-a-chip and, more concretely, to a method and an apparatus that measure quantitatively interactions between biomolecules and fluorescently labeled biomaterials and enzyme activities using the measurement of fluorescence polarization FP. The method and the apparatus of the invention provide rapid assays with minute amounts of samples using an automated device compared to conventional methods. Accordingly, the method and the apparatus of the invention can be usefully applied to the measurement of interactions between biomolecules and to the protease assays using a protein substrate.
