Described herein are binding proteins that specifically bind to human CD33, and in particular to bispecific binding proteins that specifically bind to human CD33 and human CD3. Also described herein are bispecific tandem diabodies that bind to CD33 and CD33, and their uses for immunotherapy of CD33cancers, diseases and conditions such as acute myeloid leukemia (AML).

Bispecific CD33 and CD3 binding proteins

This application describes binding proteins that specifically bind to EGFRvIII and multispecific binding proteins that specifically bind to EGFRvIII and CD3. Further described is a multispecific tandem diabody that binds to EGFRvIII and CD3. Further described are highly cytotoxic EGFRvIII/CD3 bispecific tandem diabody for recruiting T cells to specifically and potently kill several types of solid tumor cancer.

Antibody binding sites specific for EGFRvIII

The invention relates to a humanized CD3 binding site, which comprises (a) a variable heavy chain domain (VH) as depicted in SEQ ID NO:8 and a variable light chain domain (VL) as depicted in SEQ ID NO:3; or (b) a variable heavy chain domain (VH) as depicted in SEQ ID NO:9 and a variable light chain domain (VL) as depicted in SEQ ID NO:4, or c) a variable heavy chain domain (VH) as depicted in SEQ ID NO:7 and a variable light chain domain (VL) as depicted in SEQ ID NO:2; or (d) a variable heavy chain domain (VH) as depicted in SEQ ID NO:6 and a variable light chain domain (VL) as depicted in SEQ ID NO:1. The CD3 binding sites have an increased stability, while the binding affinity has been retained due to mutations at positions VH111 and VL49.
