A laser treatment device and process with controlled cooling. The device contains a cooling element with high heat conduction properties, which is transparent to the laser beam. A surface of the cooling element is held in contact with the tissue being treated while at least one other surface of the cooling element is cooled by the evaporation of a cryogenic fluid. The cooling is coordinated with the application of the laser beam so as to control the temperatures of all affected layers of tissues. In a preferred embodiment useful for removal of wrinkles and spider veins, the cooling element is a sapphire plate. A cryogenic spray cools the top surface of the plate and the bottom surface of the plate is in contact with the skin. In preferred embodiments the wavelength of the laser beam is chosen so that absorption in targeted tissue is low enough so that substantial absorption occurs throughout the targeted tissue. In a preferred embodiment for treating large spider veins with diameters in the range of 1.5 mm, Applicants use an Er:Glass laser with a wavelength of 1.54 microns.

A enhanced method for observing tumor progression, angiogenesis and/or metastasis in animal models in real time is described. The invention employs a skin flap over the area to be observed that can be opened and closed reversibly. The invention also permits simultaneous observation of more than one tumor by use of multiple colors.

The invention relates to the whole-body external optical imaging of gene expression. Specifically, methods for whole-body external optical imaging of gene expression and methods for evaluating a candidate protocol or drug for treating diseases or disorders using a fluorophore operatively linked to the promoter of a gene and external optical imaging are provided herein. Methods to screen for substances or genes that regulate target promoters are also provided.

Whole-body optical imaging of gene expression and uses thereof

The present invention provides a methods and apparatuses for use in providing rapid, selective and effective targeted delivery of beneficial substances into hair follicles. The invention relies on the use of vibrational method of delivery of substances of different size from small molecules up to particles compatible with the diameter of hair strand into the hair bulb. The skin is subjected to the vibration of about 1 Hz to about 100 Hz frequency and 0.1 mm to about 10 mm amplitude, an amount which is sufficient to cause mechanical shifts in different directions of skin structure without damage. This results in follicle effective uptake of the tested substances such as carbon particles of different size as well as melanin. The methods and apparatuses of the present invention are particularly useful in conjunction with photosensitive agents and electromagnetic irradiation. Such a combination is used to target the delivery of the agent to within hair follicles and then subsequently activate the agent.
