The present invention relates to an isolated microorganism belonging to the genus, characterized in that it comprises inactivating the gene Rv0757 that confers a PhoP&#x2212; phenotype and inactivating a second gene that prevents the production of DIM (DIM-phenotype). Additionally, the present invention comprises the use of said microorganism for producing a vaccine for immunizing against or preventing tuberculosis.

Tuberculosis vaccine

An isolated microorganism belonging to the genus, having one or both of an inactivated gene Rv0757 that confers a PhoP&#x2212; phenotype and an inactivated second gene which prevents the production of DIM (DIM&#x2212; phenotype). Methods of making the microorganism, related pharmaceutical formulations and vaccines are also provided, as are related methods of treatment and vaccination. The pharmaceutical formulation may also serve as a vector or adjuvant.

The construction of a mutant in the phoP gene by means of homologous recombination from a clinically isolatedreduces the virulence thereof in mouse bone marrow macrophage. Moreover, the phoP mutant reduces the virulence thereof in the experimental mouse model. Said phoP mutant can persist without being eliminated both in the macrophage and in the mouse. Mice inoculated with the phoP mutant are protected againstinfection. The use of mutants of mycobacteria in which the phoP gene or the genes regulated by phoP have been inactivated are candidates for vaccines against human and animal tuberculosis as well as possible recombinant vaccines against other pathogens.
