A closed system centrifuge tube is described and discussed herein that enables removal of waste and transfer and removal of samples via one or more ports of the centrifuge tube within the closed system, without exposing the sample to the outer environment. This may enable greater sample retention during sample processing and centrifugation of the sample.

A lighting system includes a light source configured to generate light to inactivate one or more pathogens. The light includes an inactivating portion. In one embodiment, a method for inactivating one or more pathogens and optionally concurrently illuminating a room having one or more human occupants while the pathogens are inactivated is also provided. The method includes generating light from a light source to inactivate the one or more pathogens. The light is generated with an inactivating portion of the light.

The present disclosure relates to approaches for assessing a sample or the presence of microorganisms. The sample, in certain implementations may be assessed for one or both of absence of microorganisms (sterility) and/or for concentration of said organisms (bio-burden). sample partition device may be employed that partitions the sample input volume into multiple discrete measurement zones with little or no loss of sample (e.g., zero-loss) and with little operator involvement, thereby reducing operator- and environment-based false positives.

Provided herein are methods for the collection and amplification of circulating nucleic acids from a non-celluar fraction of a biological sample. Circulating nucleic acids are extracted from the non-cellular fraction and are circularized to generate single-stranded nucleic acid circles, which are then subsequently amplified by rolling circular amplification using random primers to produce an amplified library. Devices for the collection of a non-cellular fraction from a biological sample are also provided. The device includes a filtration membrane and a dry solid matrix, which is in direct contact with the filtration membrane.

Method and device for collection and amplification of circulating nucleic acids

A system for isolating a target molecule from a bioprocess fluid includes a single-use disposable separation device having a plurality of perimeter-bonded layers defining one or more mesofluidic channels of the separation device, wherein each layer includes a biocompatible polymer material, wherein the separation device is configured to separate at least a portion of particles from the bioprocess fluid to generate a substantially clarified bioprocess fluid, and a chromatography system fluidically coupled at the outflow of the separation device in a configuration for further processing the clarified bioprocess fluid.

Methods for in vitro transcription and translation using a double-stranded concatemeric DNA in a eukaryotic cell-free expression system are provided. The method includes the steps of (a) contacting a double-stranded concatemeric DNA with a eukaryotic cell-free expression system, and (b) expressing a protein in vitro from the double-stranded concatemeric DNA in the eukaryotic cell-free expression system. The double-stranded concatemeric DNA includes a plurality of tandem repeat sequences. The plurality of tandem repeat sequences includes an expression sequence including a promoter, a cap-independent translation element (CITE), and an open reading frame. A final concentration of the double-stranded concatemeric DNA in the eukaryotic cell-free expression system is in a range from about 0.1 ng/&#x3bc;L to about 35 ng/&#x3bc;L. A RCA product DNA may be used as the double stranded concatemer DNA for the methods.

Cell-free protein expression using double-stranded concatameric DNA

The general illumination lighting system including a general illuminating light source configured to generate light toward one or more surfaces or materials to inactivate one or more pathogens on the one or more surfaces or materials. The one or more targeted surface illuminating light sources configured to generate targeted light toward one or more surfaces of the devices, equipment, or fixtures to inactivate one or more pathogens on the one or more surfaces. The light from the general illuminating light source and the one or more targeted surface illuminating light source including an inactivating portion having wavelengths in a range of 280 to 380 nanometers.

A system includes a bacteria culture array that includes a plurality of chambers each configured to receive a portion of a sample that includes bacteria. Each individual chamber of the plurality of chambers includes a chamber opening configured to permit access of the portion of the sample to the individual chamber. The system also includes one or more sensors configured to collect data from the individual chamber. The sensors are configured to contact the sample. Additionally, the system includes a monitoring and analysis system that includes a processor configured to receive the data from the one or more sensors at a first time and a second time, compare the data received at the second time to the data received at the first time, and identify a portion of the plurality of chambers of the bacteria culture array based on the comparing.

Systems and methods for rapidly sensing microbial metabolism

Provided herein are methods for generation and amplification of a single-stranded DNA circle in a single reaction vessel from a linear DNA without any intervening purification steps. The single-stranded DNA circle is generated via a template-independent single-stranded DNA ligation. Whole-genome amplification of linear chromosomal DNA in a single tube using ligation-assisted DNA amplification is also provided.

Ligase-assisted nucleic acid circularization and amplification

A solid substrate for the extraction, stabilization, and storage of proteins is provided. The substrate includes: a polysaccharide, such as melezitose under a substantially dry state. The substrate is configured to extract proteins from a sample and stabilize the extracted proteins in a dry format under ambient conditions for a prolonged period of time. Methods for collecting and recovering the proteins stored in the dry solid substrate are also described.

Substrates and methods for collection, stabilization and elution of biomolecules

A separation device, system and associated method are provided herein for separation of particulates form a base fluid. The separation device comprises a first microchannel comprising a fluid inlet and a mesofluidic collection chamber. The mesofluidic collection chamber has a first side and a second side, wherein the mesofluidic collection chamber is operatively coupled to the first microchannel on the first side, and wherein the mesofluidic collection chamber comprises a first fluid outlet at the second side, such that the fluid inlet, first microchannel, and first fluid outlet are in fluidic communication via the mesofluidic collection chamber.

Devices for separation of particulates, associated methods and systems

A method is provided herein, the method includes: applying a sample comprising target nucleic acids to a sample application zone of a substrate; and flowing a nucleic acid amplification reaction mixture across a length of the substrate through the sample application zone to amplify the target nucleic acid forming a nucleic acid amplification product; wherein the target nucleic acid having a first molecular weight is substantially immobilized at the sample application zone and wherein the amplification product having a second molecular weight migrates away from the sample application zone. An associated device is also provided.
