A device and method for observing an object, in particular a biological object includes a light source able to illuminate a sample. Under the effect of the illumination, the object emits back-scattered radiation that propagates to a screen, the area of which is larger than 100 cm. The projection of the back-scattered radiation onto the screen forms an image representative of the back-scattered radiation, called a scattergram. An image sensor allows an image representative of the scattergram formed on the screen to be acquired.

Device and method for observing the radiation backscattered by an object

A device and method for observing an object, in particular a biological object, includes a light source able to illuminate a sample, and a screen lying between the light source and the object. The screen includes an aperture, through which propagates the illuminating beam produced by the light source and propagating toward the screen. Under the effect of the illumination, the object emits back-scattered radiation that propagates to the screen, the area of which is preferably larger than 100 cm. The projection of the back-scattered radiation onto the screen forms an image representative of the back-scattered radiation, called a scattergram. An image sensor allows an image representative of the scattergram formed on the screen to be acquired.

Method for observing an object

The invention is an iterative method for acquiring a spectrum of a particle that is subjected to an illumination. It may in particular be a Raman spectrum. The method includes successively acquiring spectra that are what are called elementary spectra. These elementary spectra are combined to form a combined spectrum, which may be obtained by summing said elementary spectra. With each elementary spectrum is associated an acceptance criterion that is representative of a variation between said elementary spectrum and the elementary spectra acquired beforehand. Depending on this acceptance criterion, the elementary spectrum is either rejected, or accepted, in which case it is added to the combined spectrum. The invention makes it possible to guard against a degradation of the particle under the effect of an excessive exposure to said illumination.

Method for acquiring a spectrum

A system is provided for observing objects on a substrate which includes a light source that emits polarized light rectilinearly along a first direction, a holder that receives said substrate having a surface and includes objects, wherein at least one of the holder or the substrate are translucent or opaque, a detector that collects the backscattered light from the interaction between the light emitting by the light source and the objects, a polarization splitter and a quarter-wave plate wherein the polarization splitter and the quarter-wave plate are arranged so that the polarization splitter directs light towards the substrate through the quarter-wave plate, and wherein the light forms a beam and the system modifies the size of the beam. The system thus allows one to observe objects on a non-transparent substrate.
