Embodiments of the present invention are directed to a plurality of substantially spherical microspheres comprising at least one API substantially dispersed in at least one polymer and a lyoprotectant on an outside surface of the plurality of substantially spherical microspheres, wherein the plurality of substantially spherical microspheres have a D99[num] particle diameter of less than about 10 &#x3bc;m; a D90[num] circularity value of from about 0.8 to about 1.0; and comprise API in a weight of about 20 to about 40 wt. % of the polymer. Other embodiments relate to injectable compositions comprising such microspheres and methods of treating a number of conditions by administering such injectable compositions to a subject.

Pharmaceutical compositions and uses thereof

Various embodiments of an implant devices and systems are provided. The implant devices can include a mesh or implant encapsulated or otherwise coated, in whole or in part, with a film or coating. The coating or film can include methylcellulose or ethylcellulose to provide tissue regeneration as a hydrogel, while also providing a stiff polymer film when dry.

Film encapsulated pelvic implant system and method

Compositions, systems, and methods for the treatment of pelvic tissue conditions that include use of a silk fibroin-containing hydrogel composition are disclosed. The composition may include a water-soluble fibroin and a water-soluble crosslinkable polymer that can be used as a pre-formed hydrogel, and/or delivered to a pelvic tissue site and crosslinked in situ to form a hydrogel. The composition may promote tissue closure, such as vaginal wound or fistula closure; and/or enhance attachment or anchoring of a pelvic implant to a pelvic tissue. The composition may be a hydrogel that is delivered to a pelvic tissue site, formed at the site in situ, and/or associated with a portion of the implant (e.g., a mesh implant) via a tether or implant extension portion, thereby providing improved immobilization of the implant. Systems or kits that include the composition and/or a delivery needle or insertion tool are also described.

Fibroin-containing hydrogels for pelvic tissue treatment

Described are pelvic implants, systems including the implant and a delivery tool, and methods for the treatment of incontinence or prolapse. The implant has an expansion member associated with a support portion and a tensioning member along an extension portion of the implant. Systems and methods for treating incontinence or prolapse that include a composition having non-absorbable fibers of different sizes that can be applied to and set on an implant are also provided.

Systems, methods, and implants for treating prolapse or incontinence

The invention provides fluorinated polymeric articles formed a composition including a fluoropolymer. The fluoropolymer is formed using a fluorinated monomer that provides the fluoropolymer with most or all of the fluorine atoms not directly covalently attached to an atom of the fluoropolymer backbone. The fluoropolymer can also include a non-fluorinated hydrophilic monomer in a weight amount greater than the fluorinated monomer. The fluoropolymer composition also includes an ultraviolet light (UV)-reactive group capable of covalent bonding to promotes formation of the fluorinated polymeric article. The fluorinated polymeric article can be in the form of a durable lubricious coating on the surface of an implantable medical device. Coatings of the invention exhibit excellent wet/dry lubricity, durability, and controlled swelling.

Fluorinated polymers and lubricious coatings

The present invention relates compositions for delivering therapeutic agents from a medical device including an expandable and collapsible structure and methods employing them. A lipid coating including one or more fatty acids increases the amount of therapeutic agent released from the device at the delivery site. The therapeutic agent can be in a matrix including a hydrophilic polymer or an amphiphilic polymer. Release and adhesion coatings can also facilitate delivery of therapeutic agent.

Lipid coating for medical devices delivering bioactive agent

The invention provides polymers comprising pendent photochrome or near IR dye groups, as well as polymeric matrices made from these polymers, which can be used as or in association with a medical article. The polymers can be synthesized using methods that facilitate the preparation of medical articles having good biocompatibility. Exemplary polymeric matrices are in the form of lubricious coatings on medical devices, such as catheters. Visualization by irradiation of the photochrome or near IR dye can improve detection of the polymeric matrix on a device or in the body. This, in turn can improve aspects of a medical procedure, such as device insertion or matrix formation, as well as being useful for assessing the quality of the matrix.

Hydrophobic &#x3b1;(1&#x2192;4)glucopyranose polymers with enhanced degradation properties are described. Between the &#x3b1;(1&#x2192;4)glucopyranose polymeric portion and the hydrophobic portion exists a linker portion having a chemistry that facilitates degradation of the polymer. Diester and carbonate ester linker chemistries are exemplified. Biodegradable matrices can be formed from these polymers, and the matrices can be used for the preparation of implantable and injectable medical devices wherein the matrix is capable of degrading in vivo at an increased rate. Matrices including and capable of releasing a bioactive agent in vivo are also described.

Hydrophobic polysaccharides with diester- or carbonate ester-containing linkages having enhanced degradation

Hydrophobic &#x3b1;(1&#x2192;4)glucopyranose polymers useful for the preparation of implantable or injectable medical articles are described. The hydrophobic &#x3b1;(1&#x2192;4)glucopyranose polymer includes pendent groups having hydrocarbon groups and terminal amine or hydroxyl groups. Biodegradable matrices can be formed from these polymers, and the matrices can be used for the preparation of implantable and injectable medical devices wherein the matrix is capable of degrading in vivo. Matrices including and capable of releasing a bioactive agent in vivo are also described.
